Objective:To evaluate the correlation between its prognosis and clinicopathologicfactors in patients with gastrointestinal stromal tumor (GIST), and toinvestigate the expression of microRNAs affected the survival in gastrointestinalstromal tumor and their impact on the prognosis of gastrointestinal stromal tumors.Methods: We have collected182patients with gastrointestinal stromal tumors fromAffiliated Nanjing Hospital of Nanjing Medical University and AffiliatedHospital of Nantong University from January2001to July2011. These cases werere-diagnosed by pathologist and immunohistochemical methods.According to clinical data, we filtered out two cases, one with long survivsl time andone with short survival time. We applied microRNA chip of Agilent company andpreliminary screened out the differences in the expression of microRNA between long andshort survival time of GIST. And combined with microarray results and related softwareand documentation, and we make further analysis, screening out the targetmicroRNA.In182cases,162cases were included with complete clinical and pathological dataand paraffin tissue. We extracted RNA from paraffin tissues, and detected theexpression of microRNA640using TaqMan probe and special neck ring primer byreal time fluorescence quantitative polymerase chain reaction (qRT-PCR) medthod.We used statistical methods to analyze the relation between objective microRNA and clinicopathologic factors of these cases, and to analyze the relation between objectivemicroRNA and survival time and prognosis.Results: A roral of182patients were included:92men and90women, whose meanage was57.8(16-82years). The disease-specific survival rate was99%for1year,57%for3years, and32%for5years. The most commonly used method of stagingsystem according to National Institutes of Health (NIH), GISTs were divided intovery low,low,intermediate and high risk groups.Combined with microarray results and related software and documentation, wescreened out the target microRNA640influenced on survival time. The results ofqRT-PCR showed that158cases of them expressed microRNA640. And statisticalresults showed that a correlation existed between microRNA640andclinicopathologic factors, and expression of microRNA640was different betweenlong period and short period of survival time in GIST. Survival analysis revealedmicroRNA640were significant for prognosis, and microRNA640was an independentprognostic factor.Conclusion: In this study, we knew a correlation existed between microRNA640andclinicopathologic factors, and the survival time and prognosis were related tomicroRNA640, we could control microRNA640and its target genes to eliminate itsadverse effects, to reduce tumor progression and recurrence, to decrease mortality.
