ObjectiveTo investigate the effects of methyl methacrylate on cardiomyocyte apoptosis ofnewborn Sprague-Dawley rats by detecting the cell viability and apoptosis intex aftertreated by methyl methacrylate with different concentrations,and by checking theexpression of Bcl-2and Bax.MethodsCardiomyocyte was prepared from newborn rats and divided into sixgroups.Group1was defined as the control group,group2to6were treated bymethyl methacrylate with0.5、1.0、2.0、4.0、8.0mg/ml for2h respectively.CCK-8assay was used to detect cardiomyocyte viability and TUNEL cell staining forapoptosis,the expression of Bcl-2and Bax was detected by western blotting.ResultsMethyl methacrylate incubation for2h decreased the cell viability and triggeredapoptosis in a concentration dependent manner.Compared with the control group, thecell viability was significantly decreased to(71.0±6.6)%in group4、(55.3±6.0)%in group5、or(34.0±7.0)%in group6(P<0.01),theapoptosis index was evidently increased to (15.33±0.43)%in group4、(32.88±0.30)%in group5、or(65.00±0.59)%in group6(P<0.01),andgroup5showed remarkable decrease on Bcl-2expression and increase on Baxexpression. ConclusionMethyl methacrylate could promote the cardiomyocyte apoptosis.The possiblemechanism is to inhibit the expression of Bcl-2and to promote the expression of Bax.
