Objective: To study the effects of protective and therapeutic ofcurcumin on the severe acute pancreatitis (SAP) and the possible mechanismsof TLR4/NF-κB signal transduction pathways of severe acute pancreatitis ratsby detecting the AMY、TNF-,IL-6level in blood serum,observating the grossmorphology、pathology changes and the expressions of TLR4,NF--κB inpanreatic tissues.Methods:Fifty-four SD female rats, with the weight of180-250g,were evenly devided into3group randomly:Normal group(N group),severe acute pancreatitis model group(M group),curcumin-prcondition group(Cgroup).The M group were injected5%sodium Taurocholate in1.0ml/kgretrograde into pancreatic duct of the rats,and the C group injected100mg/kgcurcumin peritoneal injection before induce the model2hours, the otherprocess were same as the M group. The Pancreas of N group were just flippedand striked gently. In M and N group,40%Aqueous Solutions of PropyleneGlycol1ml/100g peritoneal injection respectively before induce the model2hours.The serum, ascites and pancreatic tissue after the operation werecollected at3h,6h,12h in the each group.The IL-6and TNF-α contents weredetected with the ELISA in each group,the AMY level detected with theautomatic biochemistry analyzer by G7-PNP,and the expression of NF-κB、TLR4and pathological change were observed with immunohistochemistry andHE staining methods. All the results were analysed by Anova with SAS8.0. Results:①The level of AMY in C group was higher than that of N goup,butlower than that of the M group(P<0.05). The number of AMY increased at3hafter the operation,and it climbed gradually in M group.②Compared with the Ngroup,the level of IL-6and TNF-in M、C group increased obviously(P<0.05).The level of IL-6and TNF-in C group decreased obviously compared withthe M group at every time point tested (P<0.05).③The pathological change andthe scores of pancreatic tissue:the pathological changes of pancreatic of Ngroup were normal in general,congestion and edema gently,little inflammationcell infiltration.The structure of pancreatic lobule of M group wereheterogeneity at every time point in M group, interlobular septum increasedobviously,the congestion edema bleeding、 necrosis inflammatory cellinfiltration had severe in the pancreatic tissue., pancreatic duct epitheliumacidophilia and lobule deletion at12h extreme obviously.the congestion、necrosis and infiltration of the C group was better than M group in each timepoint. The schimdt score of M、C group was higher than that of N group,Cgroup was lower than M group(P<0.05).④The expressions of TLR4、NF-κB:Compared with the N group and C group, the expression of NF-κB and TLR4of M group was increased obviously at each time spot, meanwhile theexpressions of them decreased obviously in the C group compared with the Mgroup. In addition,the expression of TLR4was parallel with that of NF-kB. Thelevel of expression TLR4and NF-κB was consistent with their morphologicchanges.Conclusions: 1.we succeeded in duplicating the SAP model by retrograde injected sodiumTaurocholate into panereatic duct.2.The expression of NF-κB and TLR4increased in pancreatic tissue of SAP,and it suggests that activation signaling pathway mediated by TLR4and NF-κBinvolved in the occurrence and development of SAP, and related to thepathogenesis of SAP closely, and as one of the important mechanisms thatmight damage organs distant to the pancreas and lead to pancreatic hemorrhageand necrosis.3.Curcumin could reduce the damage of SAP through suppressing theNF-κB signaling pathway activation to certain extent, and thus it might possessa protective and therapeutic effect.
