| Objective:To investigate the influence of recombinant growth hormone on tumor necrosis factor-a(TNF-a) and insulin-like growth factor-1(IGF-1)〠liver function and endotoxin on rats with obstructive jaundice.To discuss the treatment mechanism of recombinant human growth hormone in obstructive jaundice.Methods:Adult Wistar rat (any male and female)70, weight300g-350g. Randomly divided into six groups, sham operation (SO) group n=1o, sham operation with recombinant human growth hormone (rhGH SO) group n=10, obstructive jaundice (OJ) group (7days,14days,) n=13; Recombinant human growth hormone (rhGH) group (7days,14days), n=13. On postoperative7days and14days serum were harvested, To measure the ALTã€AST and endotoxin with serum. Insulin-like growth factor-1(IGF-1) and tumor necrosis factor-a(TNF-induced) was measured by Enzyme-Liked ImmunoSorbent.(ELISA) with rat serumResults:Compared with obstructive jaundice group:1. IGF-1in recombinant human growth hormone group was significantly lower than that in the obstructive jaundice group (P<0.01);2. TNF-a in recombinant human growth hormone group was significantly lower than that in the obstructive jaundice group (P<0.01);3.the concentration of ALT and AST in recombinant human growth hormone group is decreasing than that in obstructive jaundice group (P<0.01);The changes of IGF-1compared with endotoxin in Recombinant human growth hormone group becomes negative correlation (r-0.761, P<0.01).Conclusion:1. Recombinant human growth hormone can significantly reduce the damage to the liver, and prevent the raise of endotoxin in obstructive jaundice.2. Recombinant human growth hormone can interfere the expression of IGF-1and TNF-ain rats with obstructive jaundice. It have effective treatment to obstructive jaundice. |
