Quality Evaluation On Resources Of Radix Ophiopogonis

This paper studied morphological characters, pollen morphology, micromorphological characters of leaf epidermis of Ophiopogon japonicus (Thunb.) Ker-Gawl., determined the content of active components of Radix Ophiopogonis resources, and discussed the influence of soil factors on the internal quality of Radix Ophiopogonis, in order to provide the theoretical basis for the production of high quality Radix Ophiopogonis. The results were as follows:(1) Morphological characters of twelve O. japonicus germplasm materials were observed and compared. The results showed that morphological characters had significant differences among twelve germplasm materials. O. japonicus from Santai and Ya’an, Liriope spicata (Thunb.) Lour. var. prolifera Y. T. Ma from Luojiang, Liriope muscari (Decne.) Baily from Xiangcheng had high yield characters:such as the leaf was short, the root was long and wide.(2) Pollen grains and leaf epidermis micromorphology of different germplasm materials of O. japonicus. were observed and compared by the method of Scanning Electron Microscope (SEM). The results showed that the pollen grains were all prolate in shape,which had particles-reticular sculpture in pollen exine; the ditch length of far pole single groove nearly reached two ends, which was bilateral symmetry. The stomata on the leaves of O. japonicus. showed ellipse with no subsidiary cell, bogging down and zonal distribution on lower epidermis, no stomata was observed on upper epidermis. Lower and upper epidermis were rich in waxy.(3) It was detected on the content of carbohydrate, flavonoids, saponins and other active components in Radix Ophiopogonis of different populations. And indicators which required in pharmacopoeia wre measured, including water-soluble extract, ash, total ash, moisture. The obtained data were analysed with PASW statistics18.0software. And it was determined in the active components content in different parts (root crop, fibril and leaf) of Radix Ophiopogonis from different germplasm resources, and compared the distribution and accumulation of polysaccharides, flavonoids and saponins in Radix Ophiopogonis different parts. The results showed that Radix Ophiopogonis produced in Cixi and Santai were clustered in a kind, genuine medicinal materials of Radix Ophiopogonis could be distinguished with other non-genuines by composite indicators. Polysaccharides content of genuine medicinal materials of Radix Ophiopogonis from Cixi and Santai was more than other populations significantly, but their saponins content was loess than other populations significantly. Polysaccharides and total flavonoids content in root crop of Radix Ophiopogonis increased year by year. It was found that a variety of active component content in fibrous root was more than in root crop significantly.(4) It was detected on inorganic element content, physical and chemical properties from7planting soil of Radix Ophiopogonis. The corresponding contents of active components and inorganic element in Radix Ophiopogonis were also analyzed. And it was studied on the data of soil and Radix Ophiopogonis with path analysis and gray correlation. The results showed its growth soil was in neutral or weak alkaline mostly, and it regulate the content of inorganic elements primarily through the active absorption; the influential factor to the active components of Radix Ophiopogonis is the soil enzyme activity; potassium, pH and organic matter was followed by; and K, Fe, Mn, B, Ba, Zn on the impact of the quality of Radix Ophiopogonis is greater than other inorganic elements.(5) Study on residues of organochlorine pesticides and heavy metals in the four main producing areas of Radix Ophiopogonis and corresponding soils. The results showed that the residues of organochlorine pesticides in Radix ophiopogonis were lower than the permissible maximum limits of our national standard except hexachloride (BHC) in Radix ophiopogonis of Cixi. The enrichment capacity of Radix ophiopogonis for (BHC) is stronger than the DDT.
