Effects Of Bizhongxiao Decoction And The Demolition Party On P-P38、P-JNK Expression In Synovium Of Collagen-induced Arthritis Rat

Objective:Decetion of CIArats in synovial tissue mitogen-activated protein kinase pathway in P-P38and P-JNK expression and the impact of BZX (BiZhongXiao Decoction, BZXD) its decomposed formulas (wet, hot, toxic, stasis splitside) on CIA rats. Further explore the pathogenesis of the signal transduction pathways in rheumatoid arthritis (with rheumatoid arthritis, RA), to explain the mechanism of action from the wet, hot, toxic, stasis on the Treatment of RA.Methods:All rats were received subcutaneous injection of CII and CFA. Then they were randomly divided into6groups:normal contral group,model group, BZXD and the demolition party group.The model group were filled with water.Observe general condition,weight,arthritis index score,degree of paw swelling in3groups; P-P38and P-JNK were measured by western blot in synovial tissue.3.0Imagetool grayscale scanning software, semi-quantitative analysis of the results, use spss Statistical Package to analysis the results.Results:1、CIA model could be copied by BIIC and CFA.After1week of the initial immunization,the CIA rats were appear arthritis symptoms.After1week treatment, arthritis symptoms began to decline, weight gain faster, paw swelling reduced in BZXD and the demolition party group,but the model group is rising. 2、Abundant vascular proliferation,inflammatory cells and synovium incrasstion were observed in model group;but a few neonatalvasculars and inflammatory cells were observed in threpy group.3、Compared with the normal group, the expression of P-P38and P-JNK in the model group were significantly higher (p<0.05). Compared with the model group,the BZXD and demolition group could reduce more of the production of P-P38and P-JNK(p<0.05). Compared with demolition groups, the BZXD group could reduce more of the production of P-P38(p<0.05),but can not reduce more of the production of P-JNK(p>0.05).Conclusion:1、Subcutaneous injection of BIIC and CFA imitates CIA model successfully.2、During the process of CIA,P-P38、P-JNK expression were significantly enhanced, prompted that P-P38、P-JNK may be involved in the process of RA synovium and cartilage disease.3、BZXD group and demolition group could reduce the production of P-P38and P-JNK.It is proved that from the hot and humid toxic stasis treatment of RA could inhibition of bone erosion. BZXD group superior to the demolition group in turn down the expression of P38,JNK,suggest the wet,hot,toxic,stasis compound pathogenesis is the major traditional Chinese medicine pathogenesis in RA.
