To explore the possible mechanisms in treatment in cavernous venous malformation by copper needles retained.Method:â‘ From September2009to January2012,6cases with surface cavernous venous malformation were selected from the Department of Plastic and Cosmetic Surgery, The Third XiangYa Hospital of Central South University. All cases understood and agreed to their treatment (A total of12cases of the comparative samples were selected).â‘¡The copper needles with a blunt point and a circularity end, which were made of industrial copper wire with diameter of3mm were sterilized before treatment. All cases were retained12-56copper needles (average30) in the local lesions under anesthesia for1week.â‘¢All the samples were dyed in hematoxylin-eosine, after that we observed the histological and pathological changes in the vascular endothelial cells under the microscope of Nikon Eclipse80i. Via immunohistochemical techniques, we observed that the factor â…§ associated antigen located in the vascular endothelial cells, and the expression differences of P27kip1in the vascular endothelial cells before and after treatment, while computing the positive ratio. We observed the apoptosis of the vascular endothelial cells via TUNEL method and calculated the apoptosis index. We took pictures by high resolution Nikon DS and NIS-Elements AR3.0software. We used the Ver3.00software for digital image acquisition and the SPSS17.0statistical software for statistical analysis.Results:All the6cases were involved in the result analysis.â‘ After treatment, all the area of the lesions were narrowed compared with before.â‘¡Histological and pathological changes:Before treatment, part of blood vessels showed a cystic irregular expansion of its lumen sizes, wall of uneven thickening in the cavernous venous malformation; After treatment, most of the vascular structure were shrank or disappeared, while remaining a large number of lightly stained and unclear connective tissue with a lot of inflammatory cells.â‘¢The factor â…§ associated antigen was observed as dispersed brownish yellow granules in the cytoplasm of endothelial cells in cavernous venous malformation.â‘£The expression of P27kip1was significantly higher after treatment than before in nucleus of the vascular endothelial cells(P<0.05).⑤Varying degrees of apoptosis were appeared before and after treatment. The level of apoptosis was not significant when compared with before(P>0.05).Conclusion:â‘ After treatment, the size and area of the lesion were reduced. Also, the vascular lumenã€vascular endothelial cells and vascular structures were narrowed or disappeared.â‘¡The expression of P27kipl was significantly higher after treatment than before in nucleus of the vascular endothelial cells.â‘¢Varying degrees of apoptosis were appeared before and after treatment.
