Objective:This experiment is to study the a-tocopherol effect on the nerve dysfunction and morphological changes of the brain tissue, cerebral edema, Bax and Bcl-2protein expression after intracerebral hemorrhage in rats, to explore it’s mechanism of neuroprotective effect, in order to provide a theoretical basis for the clinical treatment of intracerebral hemorrhage.Methods:Weighing250~270g of healthy male SD rats of144, were randomly divided into sham operation group (n=48), intracerebral hemorrhage group (n=48) and a-Tocopherol group (n=48), each group according to6hours,1day,3days,7days4time points randomly divided into4subgroups, each subgroup12rats. a-Tocopherol group, two weeks before the operation, daily intraperitoneal injection of a-tocopherol600mg/kg, continuous injection for21days. Intracerebral hemorrhage group, sham operation group injected corresponding dose of saline at the same time. According to the Sprague Dawley rat brain atlas, in a stereotactic instrument to determine the caudate nucleus and slowly injected of aterial tail blood50ul to make rat model of intracerebral hemorrhage, sham operation group, only injected corresponding dose of physiological saline. Observing the3groups of rats’symptoms of neurological impairment at the corresponding time point after operation and make neurological function score according to Modified Neurological Severity Scores table. Rats in each group were killed at the scheduled time points, by dry and wet weight method to assay water content, brain tissue sections to observe the pathological changes, immunohistochemical method to detect the protein expression levels of Bax, Bcl-2around the hematoma in the brain tissue.Results:1. By comparative observation of brain tissue slices can be seen, that the brain tissue damage and neurological deficits of rats with a-Tocopherol treatment less than rats of intracerbral hemorrhage group.2.By dry and wet weight method, compared with rats of intracerbral hemorrhage group, a-Tocopherol can significantly reduce brain edema after intracerbral hemorrhage (P<0.05).3. Compared with rats of intracerbral hemorrhage group, in a-Tocopherol treatment group the expression of the Bax protein around the hematoma decreased (P<0.05), Bcl-2protein expression elevated (P<0.05).Conclusion:1.a-Tocopherol can alleviate the extent of the damage of nerve cells after intracerebral hemorrhage, and significantly reduce the degree of neurological deficit after intracerbral hemorrhage.2. a-Tocoph-erol plays a brain protective effect after intracerebral hemorrhage by the way of reducing brain edema.3. α-Tocopherol plays a brain protective effect after intracerebral hemorrhage by reducing the cell apoptosis after intracerebral hemorrhage through the upregulation of Bcl-2protein expression on and downregulation of Bax protein expression.
