Objective:To investigate the expression of IL-12,IL-17,TNF-αand IFN-y in colonic mucosa of patients with crohn’s disease,and observe the effects of novaferon on them to reveal the possible regulating immunological mechanism.Methods:Collected intestinal mucosa specimens from16crohn’s disease patients, the experiment set CD lesions bowel section group, CD bowel section after29days of treatment group, and16healthy for normal controls group.We clipped the mucosa of the colon by biopsy forceps. Using immunohistochemistry detection IL-12, IL-17,TNF-aand IFN-y protein expression quantity, comparing expression level difference between before treatment goup and control group, before treatment goup and after treatment goup.Results:1. the expression of IL-12in colon mucosa of before treatment goup was (10.150±1.327), was significantly higher than those in controlled goup(8.084±1.148,P<0.01), significantly higher than the after treatment goup (8.244±1.256,P<0.01).2. the expression of IL-17in colon mucosa of before treatment goup was (10.570±1.894), was significantly higher than those in controlled goup(7.250±0.729,P<0.01), significantly higher than the after treatment goup(7.362±1.079,P<0.01).3. the expression of TNF-a in colon mucosa of before treatment goup was (10.560±2.459), was significantly higher than those in controlled goup(7.258±0.703,P<0.01), significantly higher than the after treatment goup(7.424±0.392,P<0.01).4the expression of IFN-y in colon mucosa of before treatment goup was (11.010±1.732), was significantly higher than those in controlled goup(7.163±0.607,P<0.01), significantly higher than the after treatment goup(7.573±0.565,P<0.01).Conclusion:the expressions of IL-12,IL-17,TNF-α and IFN-y in mucosa of patients with crohn’s disease were increased.Novaferon could down-regulate proinflammatory cytokine,this maybe the mechanism for its treatmentof CD.
