Expression Of HMGB1and Smac In Ovarian Epithelial Tumors And Their Significance | | Posted on:2013-04-08 | Degree:Master | Type:Thesis | | Country:China | Candidate:C X Zhou | Full Text:PDF | | GTID:2234330374988612 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective:To observe the expression of HMGB1and Smacã€cell apoptosis in the tissues of epithelial ovarian cancers and analyse the two kinds of proteins’ clinical significances in the epithelial ovarian cancers.Methods:The histological diagnosis was detected by HE staining; The expression of HMGB1and Smac protein in the epithelial ovarian cancers and the normal ovarian tissues detected by the SP immunohistochemistry method; and the apoptosis in the epithelial ovarian cancers and normal ovarian tissues detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) staining method.Results:1ã€The SP immunohistochemistry method showed that the positive rate of HMGB1in the normal ovarian tissuesã€benign epithelial ovarian cancers and malignant epithelial ovarian cancers were as follows:15.00%ã€16.67%ã€66.67%, the positive rate in the malignant tumors was significantly higher than the benign tumors and normal ovarian tissues, and the difference was significant(P<0.05); but the difference was not significant in the benign tumors and normal ovarian tissues(P>0.05). And the expression of HMGB1had nothing to do with the age〠pathological typeã€histology gradeã€clinical stages of malignant epithelial ovarian cancers,but related to lymph node metastasis. The positive rate in the group of having lymph node metastasis was significant higher than the no lymph node metastasis group,and the difference was significant(p<0.05)。2.The SP immunohistochemistry method showed that the positive rate of Smac in the normal ovarian tissuesã€benign epithelial ovarian cancers and malignant epithelial ovarian cancers were as follows:80.00%ã€61.11%ã€20.00%. The positive rate in the malignant cancers was significantly lower than the benign cancers and normal ovarian tissues, and the difference was significant(P<0.05); but the difference was not significant in the benign cancers and normal ovarian tissues(P>0.05). And the expression of Smac had nothing to do with the ageã€pathological typeã€histology gradeã€clinical stages and lymph node metastasis of malignant epithelial ovarian cancers.3.The TUNEL method showed that the apoptosis index (%) in the normal ovarian tissuesã€benign epithelial ovarian cancers and malignant epithelial ovarian cancers were as follows:6.200±0.01964.520±0.0069ã€1.720±0.00359; the apoptosis index in the malignant cancers was significantly lower than the benign cancers and normal ovarian tissues, and the difference was significant(P<0.05); but the difference was not significant in the benign ovarian cancers and normal ovarian tissues(P>0.05).The apoptosis index of HMGB1positive expression malignant epithelial ovarian cancers was significant lower than the HMGB1negative expression malignant epithelial ovarian cancers,and had significant difference(P<0.05); and the relative analysis showed that the expression of HMGB1in the malignant ovarian cancers had negative correlation with apoptosis index;4. The SP immunohistochemistry method showed that in the malignant cancers, the expression of HMGB1protein had negative correlation with the expression of Smac.Conclusions:1ã€HMGB1was over-expressed in epithelial ovarian cancers, correlated with the lymph node metastasis, and had negative correlation with apoptosis index, indicated that the over-expression of HMGB1would be related to the occurrence and development of epithelial ovarian cancers by inhibition of cancer cell apoptosis; HMGB1may play a role in the metastasis of the malignant epithelial cancers;2ã€Smac was low-expressed in epithelial ovarian cancers, which indicated that the low expression of Smac would be related to the occurrence and development of epithelial ovarian cancers;3ã€The over-expression of HMGB1and low-expression of Smac maybe play an important part in the occurrence and development of epithelial ovarian cancers by incontrollable of the cell apoptosis. | | Keywords/Search Tags: | high mobility group boxl, Smac, epithelial ovariancancers, immunohistochemistry, TUNEL, cell apoptosis | PDF Full Text Request | Related items |
