[Objective] To use quantum dot fluorescence probe technology of bile duct carcinoma antigen combined ELISA for CA199testing, to explore the quantum dot technology in cholangiocarcinoma diagnosis and application.[Methods] By using synthesize with method of aqueous phase CdTe quantum dot was through modified the surface of CdTe quantum dot synthetic CdTe/CdS/ZnS into core-shell structure quantum dots. The CA199antibody conjugated quantum dots combined with a sandwich enzyme-linked immunosorbent assay (ELISA) was used to detect cancer antigen199(CA199).[Results] The synthesis get the CdTe of quantum dots with center wavelength of540nm. After the modified of CdTe/CdS/ZnS core-shell structure quantum dots had their center wavelength of630nm. Quantum dot ELISA could detect the CA199antigen at0.01ng/ml, which is100times lower than the classic ELISA. Scanning photoluminescence (PL) spectroscopy was performed on dried ELISA wells and the results were compared with the same quantum dots samples dried on a solid substrate. The "blue shift" of PL spectral was up to26nm when quantum dots conjugated to CA199antibodies. Increasing of the "blue shift" of PL spectral was observed at lower CA199antigen concentrations.[Conclusion] The CdTe quantum dots and further the CdTe/CdS/ZnS core-shell structure quantum dots have been achieved and it also verify the possibility of application of quantum dot technology and ELISA method on cancer diagnosis. This research could make the ELISA detect the antigen more easily. In experimental methodology, it provides an important idea and also a well prospect on the early diagnosis of cancer.
