[OBJECTIVE]The aim of this study is to screen the active extracts from Ipomoea batatas Lam. extracts by animal models, and to study the effects of these extracts on platelet and possible mechanism against thrombocytopenic purpura.[METHODS]These five sections of extracts were isolated from Ipomoea batatas Lam. alcohol extraction by traditional methods with different solvents, which were petroleum, chloroform, ethyl acetate, n-butyl alcohol into five polar sections.We picked out the pharmacological active section of Ipomoea batatas Lam. with the number of platelet as index by building animal thrombocytopenic models. At the same time test the level of IL-4, IFN-y of animal thrombocytopenic models in the blood of mice and study the mechanism of action initially.[RESULT]The result of the study reveal that there were phenols, organic acid, protein, glycoside, amino acid, flavonoid, saponin in the Ipomoea batatas Lam. by qualitative analysis of chemical constituent. Different solvents were used to separate the alcohol extract of Ipomoea batatas Lam. into five sections, which were petrolium section, chloroform section, ethy acetate section, n-butyl alcohol section and residual water-soluble section.Animal experiment in vivo showed that ethy acetate section, n-butyl alcohol section may raise the number of platelet. Chloroform section had rapid and significant role so we confirm it may be the effective position of Ipomoea batatas Lam. in antiplatelet reduced aspect.Chloroform section and positive control group knocked the level of IFN-γ down in the blood of mice. Chloroform section, ethy acetate section, n-butyl alcohol section and residual section could raise the level of IL-4but chloroform section and n-butyl alcohol section were significant particularly. The ratio that IFN-y to IL-4of several treatment groups reduced significantly.[CONCLUSION]This study shows there are phenols, organic acid, protein, glycoside, amino acid, flavonoid, saponin in the Ipomoea batatas Lam.Chloroform section is the effective position of Ipomoea batatas Lam. in antiplatelet reduced aspect.The state of thrombocytopenic purpura is concerned with the content of IFN-γ and IL-4.The pathogenesis of thrombocytopenic purpura may be concerned with the disorder of immune system and the abnormal expression of several cytokines.
