Objective:In the paper, the endothelial progenitor cells of the mice were isolated after cultivation. The expression of the antigen-1of the stem cells was measured. The impacts of the cigarette smoke extract (CSE) concentration on the proliferation of the endothelial progenitor cells and the expression of the antigen-1of the stem cells were studied. The study helped to discover the differentiation and the self-renewal of the mice endothelial progenitor cells under the condition of smoke exposure.Method:The cultivated long-bone marrow cells were taken from15Wistar rats of4weeks old. At the7th day, the adherent cells were taken for the identification of endothelial progenitor cells as well as the measurement of the expression of the antigen-1. The endothelial progenitor cells were divided into four groups and then placed into orifice. The endothelial progenitor cells of the control group were cultivated by original culture medium. The other3groups of endothelial progenitor cells were intervened by1%,2.5%, and5%cigarette smoke extract solution accordingly. The numbers of the living cells were measured by MTT colorimetric method after3,6, and24hour. The expression of the antigen-1of control group was compared with those of the intervention groups by Western Blot method. Results:The rats’marrow cells could be induced into endothelial progenitor cells by Density Gradient Centrifugation method and the endothelial progenitor cells could be cultivated in the EGM-2MV culture system, which was proven in the study. This study provided a promising method for the application of EPC on the disease intervention. Cigarette smoke extract (CSE) had a negative impact on the proliferation of the endothelial progenitor cells. The higher concentration of the CSE solution would lead to the stronger inhibition of the proliferation. The endothelial progenitor cells (EPC) of the rats had a high expression rate of Sca-1. However, the CSE tended to lower the expression rate of Sca-1.Conclusion:The EPC of the rats are the perfect choice for the seeds cells due to their high expression rate of Sca-1. The Cigarette smoke extract (CSE) will suppress the proliferation of the EPC as well as the expression of Sca-1. Sca-1plays a significant role in the self-renewal and differentiation of the rats’endothelial progenitor cells.
