Objective:To study the role of ACE2and FZD1in the occurrence, development and prognosis evaluation of hepatocellular carcinoma (HCC).Material and Methods:Tumor tissues from59patients with primary liver carcinoma are objects in this study, and the corresponding paracancerous tissues are for control, all samples are detected with ACE2, FZD1by Immunohis to chemical Envision two-step method. The lifetime after surgery was also analyzed.Results:1.The results of experiment shows that the positive expression of protein ACE2in HCC was57.6%(34/59), the expression in adjacent tissues is74.6%(44/59),and the normol tissues is75%(6/8); the expression in HCC is lower than adjacent tissues and normol tissues. the differences were statistically significant (χ12=8-644,χ22=9.836, P<0.05). Meanwhile, the degree of expression was not related with differentiation of HCC.2.The positive expression of protein FZD1in HCC was69.5%(41/59), which was higher than that in adjacent tissues was50.8%(30/59), the differences were statistically significant (χ2=17.11, P<0.05),At the same time, the levels of expression have negative relation with differentiation of HCC.3. the expression of protein ACE2shows significant negative correlation with the levels of protein FZD1, and the relations was statistically significant(r=-0.304, P<0.05).4.The average survival time of group with negatively and weakly positive ACE2expression was (23.29±1.48)months,the median survival time was (21±2.57) months; the average survival time of group with moderately and strongly positive ACE2expression was (21.19±2.12) months, the median survival time was (20±1.14) months, the difference between the two groups were not statistically significant. The average survival time of group with negatively and weakly positive FZD1expression was (24.81±1.79)months,the median survival time was (25±2.38) months; the average survival time of group with moderately and strongly positive FZD1expression was (20.04±1.51) months, the median survival time was (18±1.32) months, the difference between the two groups were statistically significant.5.The expression of protein ACE2have relation with cirrhosis, not gender, age, tumor emboli, tumor size, hepatitis, AFP, and the tumor metastasis. And the expression of protein FZD1was only related with tumor metastasis.6. Multivariate anarysis showed that the differentiation degree, FZD1expression were independent prognosic factors to HCC.Conclusions:1. Protein ACE2has lower expression in HCC, related with cirrhosis; Protein FZD1have higher expression, related with tumor metastasis ACE2and FZD1participated in the development of HCC. 2. The expression of FZD1have negative relation with differentiation of HCC, and lower expression suggest a well biological behavior.3. The expression of protein ACE2shows significant negative correlation with the levels of protein FZDl,they may have reversed action in the development of HCC.4.The different levels of protein FZD1have influence on the survival time of the patients. FZD1is likely to be a valuable indication in evaluation of the prognosis of patients with HCC.
