ObjectiveStudy of the impact on the express of NF-KB and HIF-1a in rat’s myo-cardial cell with Mild Hypothermia therapeutic after CPRMethodsChoose twenty SD rats, Asphyxiation by clamping the trachea was used to make the mode of CPR. The rats were randomly divided into four groups:Sham operation group, room temperature treated group,34℃treated group,32℃treated group. The sham operation group was treated by Catheterization of Arterial and venous, tracheal intubation, and Take the myocardial tissue samples6hours later.the other groups Take the myocardial tissue samples6hours afer CPR. Western-bloting was use to test HIF-1a and NF-KB protein expression of each group’s myocardial tissue. The optical microscopy is used to observe the injury of myocardial tissue.Results1. The expression of HIF-1a and NF-KB protein in room temperature treated group,34℃treated group and32℃treated group are signigicantly higher than in sham operation group (P<0.01)2. The expression of HIF-1a and NF-KB protein in34℃treated group and32℃treated group are signigicantly lower than room temperature treated group (P<0.05)3. The expression of NF-KB protein in32℃treated group is signigicantly lower than and34℃treated group (P<0.05), but The expression of HIF-1a protein in32℃treated group isn’t signigicantly lower than and34℃treated group (P>0.05) 4. The HE stain showed the injury of myocardial tissue after CPR,each group has various degree of Inflammatory reaction and necrosis,but the injury of myocardial tissue in Mild Hypothermia therapeutic group is lighter than in room temperature treated group.ConclusionMild Hypothermia therapeutic can protect the myocard after CPR through decreasing the the expression of HIF-1a and NF-KB protein. The hypothermia of32℃play a better protective role in the injury of reperfusion than34℃,but it increased the side effect such as amyos-tasia,which should be taken seriously.
