Objective:The purpose of this study was to fabricate an three-dimensional nerve conduit who itimates nerve bundle tube with related technologies, using a macaque sciatic nerve as model and polyglyolic-lactic acid (PLGA)as material.Methods:A certain segment of macaque sciatic nerves is resected, the image of excision can be observed by continuous ultramicrotome and hematoxylin-eosin[H&E]and immunohistochemical staining, as a result we can get a complete picture of each layer under slice optical microscope. And then all pictures were input into the computer in turn, and three-dimensional reconstruction images are made through E-Feature software, obtaining three-dimensional moving image of the defect nerve bundle of the whole nerve. Then we make3D nerve conduit including porous multi-cavity and multi-barrel pipe bending system.Results:The three-dimensional reconstruction of nerve conduit was successfully made by computer digital system just like the sciatic nerve of macaque.Metal filaments can be casted into curve model of three-dimensional nerve conduit by pulling and dialing technique.PLGA(PLA:PGA=85:15)can be used to make3D nerve conduit with porous multi-cavity and multi-barrel.Conclusions:An artificial nerve conduits with a certain aperture and porosity in wall, simulating three-dimensional moving of nerve bundle was successfully made, which give basis for further study of bionic artificial nerve conduits complex with NGF、BDNF、NT-3and Schwann cell.
