Purpose Discussion the function of survinin and caspase-3in rats with manifestal spina bifida.Methods put60adult female SD rats and adult male SD rats togerther in cages, according to the proportion of3:1, the next morning to do vaginal smears,the rats which snears has sperms as Pregnant rats with zero day gestation, and there are45rats have successful pregnancy. In pregnancy10days,the female rats are randomly divided into experimental group, control group and normal group. The experimental group are filled with all-trans retinoic acid via gastric tube according to135mg/kg dose, the control group was given the same volume of olive oil, normal group without any treatment. The experimental group, control group and normal group are again randomly divided into the15days,17days and19days groups, and take out the embryos with laparotomy respectively in pregnancy15days,17days,19days. Each group randomly selected20fetuses, randomly selected10of them for RT-PCR detection the expression of survinin and caspase-3,10for TUNEL staining for detection of the apoptotic cells surrounding the lumbosacral neural tube.Results1. In pregnancy15days,17days,19days,The spina bifida malformation rate of experimental group is respectively42.17%,56.14%,37.04%, the embryos of the control group and normal group of are all normal.2. The comparisons of apoptotic index surrounding Lumbosacral neural tube between the experimental group and the control group and between the experimental group and normal group have statistical significance (P<0.05), The comparisons in the same pregnancy between the normal group and the control group show no significant differences (P>0.05), the experimental group have statistical significance between different pregnancy (P<0.05). in the control group and normal group, The comparison between15days and17days and between17days and19days show significant differences (P <0.05). 3. The comparisons of caspase-3expression between the experimental group and the control group and between the experimental group and normal group have statistical significance (P<0.05), The comparisons in the same pregnancy between the normal group and the control group show no significant differences (P>0.05), the experimental group have statistical significance between different pregnancy (P <0.05). in the control group and normal group, The comparison between15days and17days and between17days and19days show significant differences (P<0.05).4. The comparisons of Survivin expression between the experimental group and the control group and between the experimental group and normal group have statistical significance (P<0.05), The comparisons in the same pregnancy between the normal group and the control group show no significant differences (P>0.05), the experimental group have statistical significance between different pregnancy (P <0.05). in the control group and normal group, The comparison between15days and17days show no significant differences (P>0.05), The comparison between17days and19days show significant differences (P<0.05).5. The relationship is closely between the Lumbosacral apoptosis index and spina bifida.They curves are have same direction.6. In the experimental group, the relevance between caspase-3and Survinin shows negative correlation and there is statistical significance(r=-0.890, P=0.000). Conclusions1. Retinoic acid can interfere with the development of the neural tube, resulting in spina bifida.2. The relationship is closely between the Lumbosacral apoptosis index and spina bifida.3. because survinin inhibit the expression of caspase-3, it can protect cells excessive apoptosis, and it is important to repair spina bifida, can reduce the incidence of spina bifida.
