Objective: To establish the expression pattern recognition system of the Anger‐out and Anger‐in experimental macaques to rhesus new tools for the evaluation of monkey model; To complete the establishment and evaluation of anger‐out, anger‐in emotion and induced PMS Liver‐qi invasion, Liver‐qi depression rhesus monkey models and evidence to the contrary pattern recognition system, and then provide an appropriate platform for studing pathogenic mechanism of anger‐out and anger‐in emotion; Application the expression pattern recognition system of the Anger‐out and Anger‐in experimental macaques to quantify emotional changes of monkeys, to collect blood samples and brain neurotransmitters central indicators of macaques for testing. By means of mathematical modeling to build macro and micro indicators equations to probe against the micro‐mechanism of facial expressions and PMS Liver‐qi invasion, Liver‐qi depression two incidence and drug intervention.Methods: To collect large quantities of monkeys’ expressions around national and hire experts to rate the expressions. To establish monkeys’ face database then build a monkey facial pattern recognition system, and based on "the female monkey mood rating scale experiment to establish evaluation criteria. Experimental macaques were induced to show up emotional reaction with social level stress method, then anger‐in, anger‐out emotion macaques were screened by Emotional Evaluation Scale about Female Experiment Macaque scoring, based on mentioned above experimental macaques were induced to show up PMS Liver‐qi invasion and Liver‐qi depression appearance combined with timing extrusion modeling, soon after their facial expression and behavior was secondary evaluated by scale scoring and pattern recognition, eventually Traditional Chinese Medicine Baixiangdan and shuyujiaonang were respectively used to interfere. Serum of different periods were collected, then it was detected with different brain regions of norepinephrine, dopamine,5‐hydroxytryptamine and γ‐Aminobutyric Acid (only in brain regions) content, then the results combined with scale scoring were used to evaluate the models and to verify the pattern recognition system and also discuss the Microscopic Mechanism of the the Baixiang Dan capsules and Shu Yu capsules. To do the curve fitting of serum markers and their expression points after the availability of the pattern recognition system has been determined to reveal the relationship between them.Results:1、 The expression pattern recognition system of the Anger‐out and Anger‐in experimental macaques can good identify facial expressions of monkeys and its score consistent with expert score. And score with expert score similarity is0.9592、The anger and depression factor changes before and after model are comply with law which have been evaluated by scale. And the rise and fall of various types of micro‐indicators before and after the model of PMS macaque model are in line with the performance. Use the expression pattern recognition system of the Anger‐out and Anger‐in experimental macaques to evaluate the monkeys’ expression and the follow results were found: Anger reaction and PMS liver‐qi inverse macaque anger expression scores were significantly higher (P <0.01) than control, PMS liver‐qi inverse group was significantly higher than the anger group (P <0.05), macaque PMS liver‐qi depress depression expression score was significantly higher than the control group and depression emotional response group (P <0.01) the Baixiang Dan, and Shu Yu capsule can significantly reduce PMS liver inverse, macaque anger of depression, depression score (P <0.01). The results are consistent with other two means of evaluation.3、The micro indicators and their facial expressions score of monkeys at the same period is more obvious non‐linear relationship. And the relationship between facial expression scores and each index are (X represents the facial expression score and Y is the micro level indicators here): NE: Y=8.044+0.035X+0.003X~2‐(1.692E‐5)*X~3, R~2=0.921; DA: Y=28.371+0.095X+0.009X~2‐(5.689E‐5)*X~3, R~2=0.575;5‐HT: Y=21.843‐0.203X+0.005X~2‐(2.244E‐5)*X~3, R~2=0.716. While we can see that the fit of the curve equation of the micro indicators and facial expression score reached a can be applied level of statistical by R~2.Conclusions:Macaque facial pattern recognition system can recognize monkey facial expressions good, and it could be a new an objective evaluation method for monkey model by comparing with the results of subjective observations, scale and micro‐indicators in evaluating model that was modeled by social level pressure stress. Rhesus monkeys could be induced to show anger‐out and anger‐in emotion with social level stress method, subjective observation, scales scoring and neurotransmitters detection could properly quantitate anger‐out, anger‐in emotion and symptoms of the disease. Anger‐out and anger‐in emotion had an relationship with the change of monoamine neurotransmitters content in serum, pathogenesis of PMS Liver‐qi invasion and Liver‐qi depression was closely related to monoamine neurotransmitters in serum and brain and GABA in brain. The regulation of neurotransmitters in serum and brain of Baixiangdan and Shuyujiaonang may be the mechanisms of treating PMS Liver‐qi invasion and Liver‐qi depression. And there’s a closely relationship between the facial expressions and the serum neurotransmitter that seen through the results of curve fitting.
