Objective:To systematically study the pharmacognostical of Bailongtou from Shandong province, provide experimental data and scientific evidence for establishing the quality standards.Methods:First, We from the original plant shape, raw material for medicine shape, the microdistinction, the physics and chemistry and so on several aspects have done the pharmacognosy research of Shandong province, and use the method of molecular biology to the identification of Bailongtou.Second,Determination study of the content of extracts, moisture, ash of Bailongtou referred datas to the Chinese Pharmacopeoia in2010.Third,Assaying of total flavonoids in Bailongtou from Shandong, is to establish a method for the assaying of total flavonoids in the extracts of Bailongtou and to provide basis for the study of its internal quality.Forth,Though the Liebermann-Burchard colorimetric method to quantify the content of total sterols in Pterocypsela plants of Shandong province.As well as though the UPLC-MS/MS method accurate detect the content of β-sitosterol and stigmasterol. Also contrasting the difference of content in baseleaf,stemLeaf and root.Results:First, They had obvious difference in exterior character, the shape and size of leaf, the shape and the transverse section characteristics of root. The rDNA-ITS sequence of Bailongtou can provided the molecular biology basis for the identification of different kinds of plants.Second, In the Bailongtou the average of moisture content was8.45%; the average of total ashes was.20%, the average of acid endurable ashes was1.27%;the average of water-soluble extract in cold water was18.30%,that in hot water23.60%;the average of eranol-soluble extracts was22.11%;the average of ether-soluble extracts was 3.77%.Third, The optimum extraction technology was that30-fold and70%ethanol,30minutes of reflux,and2times of ultrasonic wave extraction.The content of flavonoids in root of Pterocypsela. laciniata was1.0150%, Pterocypsela.indica was0.8965%, Pterocypsela. raddeana was0.7865%.Forth,Total sterol and two kinds of effective component content from high to low were baseleaf,stemleaf and root.Conclusions:First,The material morphology, microscopic characters and ITS sequence of Bailongtou in Shandong province had obvious difference, can be used as the basis for the variety identification.Second, The content of moisture, ashes and extract of Bailongtou in Shandong province had some difference.Three,The content of total flavonoids had difference in different sources Bailongtou of Shandong Provice.Four, The content of total sterols,β-sitosterol and stigmasterol had large distinction in different parts of Shandong Pterocypsela plants.
