The Study On The Expression Of Stem Cell Factor During The Repair Process Of The Mandibular Defect In Rats

ObjectiveTo detect the expression of stem cell factor (SCF) during the repair process of rats mandibular critical size bone defect (CSD) and self-healing bone defect,and the expression of bone marrow stromal cells in the two kinds of bone defects,so that to discover SCF plays an important role in bone defect repair.Method①To construct CSD and self-healing bone defect models in Specified Pathogen Free SD rats mandibular respectively. After the operation, the animals were sacrificed on1,3,5,7,14,21d;②Take out the model areas from the animal body, remove the surrounding mucle and fascia and such on organization, fixed the model area in10%formalin for12hours, and then put it in10%EDTA to take off the ore at4℃. Put the area into paraffin slice after the completion of the demineralization, to detect expression of SCF and BMSCs in bone defect area with immunohistochemistry;③Take out the model areas from the animal body, remove the surrounding mucle and fascia and such on organization, and put it in the-20℃freezer for precooling30minutes directly, then in the-80℃freezer frozen for24hours, after that, extract the protein from the bone defect area, the protein expression of SCF was tested using Western blot and do half quantitative analysis.Result The CSD and self-healing bone defect models have been successfully made in SD rats mandibular. The result of immunohistochemistry showed that the SCF expression was detected in the specimens of CSD and self-healing bone defect. However, the expression of SCF in CSD was less than that of self-healing bone defect. Furthermore, this was significant diffierence between the two kinds of bone defect, and BMSCs also express during the healing process in bone defect. Besides, the results of Western blot also confirmed the results of immunohistochemistry.ConclusionIn CSD and self-healing bone defect, there are the expression of the SCF, so does BMSCs. That is to say, there are may be some relationship between BMSCs and SCF in bone defect. SCF plays an important role in bone defect repair.
