ObjectiveResearch Citrobacter freundii on the carbapenem antibiotic resistance mechanismsMaterials and methodsCollection of the First Affiliated Hospital of Anhui Medical University fromJanuary2010to October2011, inpatient and outpatient submission of61specimens(excluding repeat isolates) Reference to the American Society of Clinical andLaboratory Standards Institute (CLSI) detected by agar dilution method Fraunhofer toCitrobacter minimum inhibitory concentration (MIC) of imipenem and meropenem, aswell as other common drugs. Phenotypic detection of the modified Hodge testCitrobacter freundii61carbon, isoelectric aggregation electrophoresis analysis of itsbeta-lactamase type, polymerase chain reaction (PCR) and DNA sequence detection ofbeta-lactamase genotype joint test of the resistance plasmid transfer.ResultOnly a carbapenem antibiotic resistant Citrobacter freundii modified Hodge testresults were positive,Citrobacter freundii the MIC of imipenem and meropenem are64μg/ml, were resistant to penicillins, cephalosporins, cefoxitin, aztreonam Southaminoglycoside. Transfer bonding results show that resistance to imipenem and mero-penem by plasmid transfer. Peer focusing electrophoresis showed that the Fraunhofercitrate has about pI5.2,6.0,6.7,8.0four kinds of β-lactamase, the zygote somelactamase pI6.7β-PCR detection and sequence analysis show that the bacteria produceKPC has-2gene.ConclusionKPC-2carbapenem-ene enzyme may be caused by Citrobacter freundii carbon carbapenem antibiotic resistance of the main reasons, and the resistance genes throughplasmid transfer...
