Almost one-third of the world’s population have been infected by Mycobacterium tuberculosis (MTB), the pathogen of TB (tuberculosis). In recent years, however, the emerge of multiple-drug resistant (MDR) MTB strains pose new challenges for the treatment of TB. How to find and where to find new drugs are the key issues of development of new anti-TB drugs. MTB belongs to actinomycetes. From the view point of biological evolution, we can readily speculate that other actinomycetes in the same taxonomic status of MTB are very likely to get the ability of producing anti-MTB secondary metabolites during the long-term competition.In this dissertation, I studied anti-MTB constituents of two strains of endophytic streptomyces, the cyclopeptide WAP-8294A2of Lysobacter enzymogenes OH11, and sencondary metabolites of two strains of endophytic fungi.The endophytic Streptomyces sp. LC18of mangrove plants was cultured in a10L ISP3solid medium. Four ansamycins were isolated and elucidated, including three octaketide ansamycins, namely herbimycin A (A-1), herbimycin C (A-2) and heronamycin A (A-4), a benzothiazine ansamycin, and one nonaketide ansamycin hygrocin A derivative6(A-3). Their structures were elucidated on the basis of NMR and MS spectroscopic analysis.The endophytic Streptomyces sp. AR1of cockroach was cultured in a10L Gause’s No.1synthetic medium. Three compounds were isolated and elucidated, including naphthoquinone WS-5995C (R-1),3,5-dihydroxyacetophenone (R-2),3,5-dihydroxybenzoic acid (R-3).The strain L. enzymogenes OH11was cultured in a10L liquid medium of soybean cake powder. The cyclopeptide WAP-8294A2(MJ-Z-12) were isolated and elucidated.The fungal strain F070624009(Marasmius cladophyllus) was cultured in a4L potato-dextrose agar medium. Seven sesquiterpenes were isolated and elucidated, including one new tremulane-type sesquiterpene (MJ-1), named as Cladophyllol, one new drimane-type sesquiterpene (MJ-2), named as drimane-3,8,11,12-tetraol, two new dimeric drimane-type sesquiterpenes (MJ-3and MJ-4), named as cryptoporic acids J and K, plus three known dimeric drimane sesquiterpenes cryptoporic acids D (MJ-5) and E (MJ-6), and15-hydroxyl cryptoporic acid H (MJ-7).The endophytic fungal strain F0182of masson pine was cultured in a500mL potato-dextrose agar smedium. Fusaric acid (MJ-D-1) was isolated and elucidated.
