Sensitive To Chemotherapy Effect Of Emodin On Human Ovarian Cancer Cells In Vitro And In Vivo:Roles Of Apoptotic Cell Death And SKOV3Tumor Xenografts Model

Objective:1)Effect of emodin on ovarian cancer SKOV3xenograft in nude mice;2)emodin combined with cisplatin effect on ovarian cancer SKOV3xenograft in nude miceMethods:SKOV3cells were subcutaneously injected into the left chest of nude mice. The nude mice were cultured according to the regulation of SPF grade. About10days after SKOV3cells injection, then randomly divided into four groups, treated with different concentration of emodin. Emodin was intraperitoneal injection every other day, lasting for two weeks. Evaluate the anti-tumor effects of emodin by monitoring the tumor volume and measuring the tumor weight in the end(RTV,RTV=V/V0).After the treatment was finished, Western blot analysis was used to examine the expression of associated proteins in the tumor tissues of nude mice.SKOV3cells were subcutaneously injected into the left chest of nude mice. Evaluate the anti-tumor effects of emodin by monitoring the tumor volume and measuring the tumor weight in the end(RTV,RTV=V/V0).After the treatment was finished, Western blot analysis was used to examine the expression of associated proteins in the tumor tissues of nude mice.Results:1)In the end of the treatment, the average RTV of xenografts was as follows:5.23±0.24、3.38±0.17、2.17±0.31was decreased significantly to compare groups (RTV8.45±0.47))(P<0.05).2)In the end of the treatment, the average RTV of xenografts was as follows:3.32±0.09、3.54±0.17、1.86±0.25was decreased significantly to scramble groups (RTV8.79±0.32))(P<0.05).3) The expression of caspase-3,caspase-9and survivin in the xenograft tissues of nude mice:Compared with compare group, the protein expression level of caspase-3,caspase-9increased significantly with the increase dosage of emodin, while the protein expression level of surviving indecreased significantly.4) The expression of caspase-3,caspase-9and survivin in the xenograft tissues of nude mice:Compared with compare group, emodin group, cisplatin group, emodin combined cisplatin group, the protein expression level of caspase-3,caspase-9increased significantly, while the protein expression level of surviving indecreased significantly.Conclusions:1) Different dosage of emodin effect on the xenograft nude mice, and curative effect increased with the increase dosage of emodin,while the toxicity do not increase.2)Emodin could sensitize the xenograft nude mice to cisplatin.
