Abstract/Summary:
Objective:To explore mouse embryo osteoblast cell lines MC3T3-E1cell proliferation and activity to in vitro, with further defined the molecular biology mechanism of estrogen in the reconstruction of bone.Methods:MC3T3-E1cells are inoculated in10-10mol/L,10-9mol/L,10-gmol/L,10-7mol/L17-B estradiol culture medium forl day,3days,5days and7days respectively, then to test cell proliferation capacity and cell activity through ALP and MTT, and to determine the best density and best effect time of estrogen and apply gene expression profile testing to explore the influence of estrogen in bone remolding signaling pathway and further study the molecular mechanism.Results:The results of MTT and ALP showed the ability of cell proliferation and cell activity of mouse embryos osteoblast cell lines MC3T3-E1increased apparently with estrogen.The results of microarray showed the estrogen group and control group differentially expressed genes as many as5403, up expression is1996and down expression is3407.Conclusion:Estrogen can improve the proliferation ability and enhance its cell active role of mouse embryos osteoblast cell lines MC3T3-E1cells, The patters of gene expression of the estrogen group are obviously different compared with the control group.
