Objective The study focuses on the preventive and therapeutic effect of monomeric triptolide upon rheumatoid arthritis (RA) by using of Adjuvant Arthritis(AA) models; aims to observe whether the ankle joint synovial tissue and the level of cytokines would change, discusses growth factor TNF-a, IL-1B on RA synovial angiogenesis of suppression mechanisms and to investigate the relationship between the role of anti-RA and antiangiogenesis.Methods The30Wistar rats weight about200±20g were involved, taking6as normal control group, taking24to accept injection of0.1ml Freund’s complete adjuvant in the right rear paw. The models were established after18days, and were divided into control group and treatment group, each group was12rats. Procedures were as follows:(1) normal control group:6rats.(2) treatment group:12rats.Injected triptolide0.4mg/kg into the right hind leg per rat.(3) control group:12rats. Each was injected with the same volume of sodium chloride injection, about9.0g/L,in the right hind leg.Each group was kept in the same condition, free feeding, drinking. The temperature was controlled between18-22℃. All the rats were killed after injecting for14days.Taked the ankle joint tissue, using HE staining, producing biopsy. Observed the pathological changes under optical microscope:synovial hyperplasia, inflammatory cell infiltration, cartilage bone destruction, joints surrounding tissue edema,etc. Determinated the levels of TNF-α、IL-1β in the synovial tissue of rats.Results The articular swelling degree and arthritis index of rats in triptolide group, was significantly lower than the control group (P<0.05). The synovial cells of pathological changes, synovial vascularity, vessel wall and the surrounding inflammatory cell infiltration and of rats in triptolide group was reduced. It showed that positive cells in triptolide group was significantly lower than the control group under the microscope.(P<0.05).Compare to the control group and the normal control group, the levels of TNF-α、IL-1β in the triptolide group were significantly lower(P<0.05). Conclusion Triptolide can significantly reduce the joint swelling degree and arthritis index of AA rats, improve the tolerance of rat in rheumatoid arthritis.Triptolide can significantly decrease the levels of TNF-α、IL-1β in the synovial tissue of rat, it was showed that triptolide inhibit rats synovial angiogenesis mechanisms was related with the expression of TNF-α、IL-1β in the synovial tissue.
