Objective:To investigate the influence of COX-2small hairpin RNA on the proliferation andthe expression of genes in HSC-T6cell.Methods:1、The cultured HSC-T6were divided into three groups: blank group, HK plasmidcontrol group,COX-2shRNA1group.2、Transfer the constructed pYr-1.1-hU6-EGFP-COX-2shRNA1into HSC-T6cellline, observe the transfection efficiency by fluorescence microscope.3、MTT were used to observe the effect of COX-2shRNA1on the proliferation ofcultured cells(HSC-T6).4、The27K Rat Genome Array from Beijing CapitalBio Corporation to findingdifferent expressed genes of COX-2shRNA1control group and HK plasmidcontrol group.Results:1、COX-2shRNA1transfected HSC after24h,48h,72h, we can see green fluorescencecytoplasm in HSC by fluorescence microscope,and the transfection efficiency ofthree time periods are over70%.2、The OD490light absorption values measured and the growth curve drawn by MTTexperiment shows the light absorption values of COX-2shRNA1group weresignificant differences after24h,48h,72h (P <0.05), the light absorption valuesof COX-2shRNA1group were statistically significant differences (P <0.05)compared with HK plasmid control group and blank group after72h.3、Transfer the pYr-1.1-hU6-EGFP-COX-2shRNA1into HSC-T6cell line,differentexpressed genes related to proliferation were detected after48h,72h,and there areeight genes,namely,Trp53,Il6,Cxcl10,Atf3, Cdc27,Csf2, Dusp1,Serpinb2. Conclusions:1、COX-2-targeting shRNA inhibits the proliferation of HSC-T6.2、The inhibition effect of COX-2shRNA1on the HSC-T6cells proliferation maybe attributable to the regulation of proliferation-related genes Trp53,Il6,Cxcl10,Atf3,Cdc27,Csf2,Dusp1,Serpinb2.
