ObjectiveThe purpose of the present study was to determine the effects of the basicfibroblast growth factor-poly lactic acid-glycolic acid copolymer(bFGF)-[poly(lactic-co-glycolic-acid))](PLGA)-sustained release microspheres(MS)by W/O/Wmultiple emulsion evaporation method on survival of a cross-area flap on the Sideabdominal of rabbits.MethodsChoose healthy Chinese Twenty-four rabbits, white body color, male and femaleunlimited, weighing1.5~2.0kg, rabbit age4months. Rabbits were randomly dividedinto three groups, each group of eight, according to rabbit Angioma median boundedsymmetric distribution before and after to the trunk. Lateral thoraco-abdominalthoracodorsal artery in rabbits and design a kiss between contacts of the iliolumbarartery size is8x12cm oval across-area flap. Preoperative5days respectively,intradermal injection were bFGF-PLGA(I group), bFGF solution orbFGF-impregnated PLGA (II group) and normal saline (Ⅲ group). After the surgery,①after7days calculated flap survival area;②common tissue slice HE staining;③Rabbit skin specimens were CD34+immunohistochemical staining to detect CD34+expression and the middle region of the flap average number of blood vessels.ResultsSustained-release microsphere group, blank microspheres and normal salinegroups after7days the average size of the flap were90.38±1.55%,61.03±1.76%,60.52±2.35%, sustained-release microsphere group survival rate was significantlyhigher than blank microsphere group and normal saline group(p<0.05), blankmicrosphere group and normal saline group difference was not statistically significant(p>0.05).Granulation tissue hyperplasia in the flap of the sustained releasemicrosphere group; CD34+immunohistochemistry showed:3groups flap among the regional average number of blood vessels, respectively (37.75±1.48)、(22.88±1.88)、(23.39±2.6)a/cm2, sustained-release microsphere group was significantly higher thanaverage number of empty vessels microsphere group and the saline group, withsignificant differences(p<0.05). Shows a lot of formation of neovascularization,bFGF-PLGA between distal blood supply, improve the survival of flaps, and moreabundant expression of CD34+.Conclusion1. The present study, just a subcutaneous local injections of preoperativebFGF-PLGA, reduce the number of dosage, improve administration efficiency.2. Preoperative5d local injection of bFGF-PLGA sustained-release microspheresin rabbit cross-area flap, it can increase flap CD34+expression, and promote flapangiogenesis, increased flap survival area. It can change the traditional methods ofsurgical delay operation, for cross-area flap grafting shorten the period of treatmentprovides experiment basis.
