Objective: Study the effect of emodin on the secretion of nitric oxide(NO)under high glucose in human umbilical vein endothelial cells(HUVECS) and disscusswhether PI3K/AKT signaling pathway is invovled.Methods: HUVECs were culured and MTT was used to screen theexperimental concentration of emodin;②Then HUVECs were randomly divided intonormal control group (group NG,5.5mmol/L glucose concentration), highconcentration group (group HG, glucose11.1mmol/L,22.2mmol/L,33.3mmol/L),the nitrate reductase assay was used to detect the secretion of NO in each group;③HUVECs were cultured with33.3mmol/L glucose concentration at different time(0,24,48,72hours),and also nitrate reductase assay was used to detect the secretion ofNO;④N ext the HUVECs were randomly divided into High glucose content group(group HG,33.3mmol/L), high glucose+emodin group, high glucose+LY294002group, high glucose++LY294002+emodin group,the same methods was used todetect the secretion of NO⑤Western blot was used to detect the expression of p-Akt(Ser473) protein of each group.Results:①HUVECs grew well with the concentration of10μ mol/L emodin(p<0.05);②Compared to the other group,the NO values were increased significantlyunder nomal glucose group (p<0.05);③Cultured HUVECs with33.3mmol/L group,as compared with0h group,24h group and NO secretion increases (p<0.05),48h,72hreduction (p<0.05);④33.3mmol/L+LY294002group compared to the other groupthe secretion of NO decreased significantly (p<0.05); emodin can improveinhibition of LY294002,emodin group NO values were compared to the other groupincreased significantly (p<0.05);⑤In the same glucose concertration,the expressin of p-Akt in emodin group is more than the other groups (p<0.05);the expressin of p-Aktprotein in Emodin+LY294002group increased significantly than LY294002groups(p<0.05)Conclusion:①HUVECs was cultured in different glucose concertration after48h,with increasing glucose concentration,NO secretion decreased.②High glucoseeffect the secretion of NO,emodin improve NO secretion under the high glucose③Emodin may to regulat secretion of ON under the environment of high glucosethrough Akt signaling pathways.
