Objective:1. To observe the effect of Endostar on MVD and MLD of the rat popliteal fossametastatic lymph node,and discuss its functions in the metastasis of the Walker-256transplanted tumor in popliteal fossa lymph nodes.2. To observe the double labeling immunohistochemistry of MVD and MLD inrat popliteal fossa lymph nodes and human metastastic axillary node of breast cancer, andexplore the relationship between lymphatic metastasis and blood metastasis.3. To observe the functions of ligation on the lymphitic metastasis in the ratWalker-256implanted tumor in popliteal lymph nodes by the ligation of the efferentlymphatics and both lymphtic and vasculars respectively.Methods:1.30SD rats were selected and randomly divided into3groups.Cells ofWalker-256was injected in the two hind pelmas of rats in group2and3to build atransplanted tumor and metastatic lymph node model. After1week when thetransplanted tumor alived and lymph nodes enlargement, Endostar0.75mg/Kg wasinjected intraperitoneally in group1,and normal saline in group2,which lasted4weeks.1week after medication finished HE staining and immunohistochemical staining were usedin popliteal fossa lymph node of the rats in3groups to calculate the metastatic rate andthe MVD and MLD.2. D2-40and CD34double labeling immunohisto chemical staining was used inboth normal and metastatic lymph nodes.At the same time30cases of breast cancermetastatic axillary lymph node specimens were selected and underwent the D2-40andCD34double labeling immunohistochemistry.3.20SD rats were selected and Walker-256cells were injected in their rightpelma to establish the transplanted tumor and metastatic lymph node model,which was randomly divided into2groups(group4,group5) after10days. Efferent lymphatics wasligated in group4and the group5was sham operative group.Popliteal fossa lymph nodesand lung tissue underwent the HE staining to observe the metastatic rate after6weeks.4. Selected20SD rats, Cells of Walker-256was injected in the two hind pelmas ofrats in group1and2to build a transplanted tumor and metastatic lymph node model..after two weeks, randomly divided into2groups(group6,group7), Foot lesions wasremoved and vascular and lymphatic ligation in group6.In group7amputation removalof foot lesions, clear the popliteal fossa lymph node. HE staining of the two sets ofstumps extremities after8weeks.Results:1. The metastatic rate of rat Walker-256transplanted tumor of popliteal fossalymph node was respectively70%and90%.(p<0.05); The MVD of the normal rat lymphnodes, the Walker-256metastatic lymph nodes,the metastasis lymph nodes Endostarintervented were respectively5.56±0.79,12.33±2.10,19.65±3.10, which got statisticalsignificance (p<0.05),The MVD were respectively6.46±2.20,10.45±2.76,12.39±2.20,which got statistical significance (p<0.05).2. The lymphatic vessels, vascular anastomosis rates within the normal rat lymphnodes, the Walker-256metastatic lymph nodes, and the metastasis lymph nodes Endostarintervented were0%,25%,8.3%,which got statistical significance(p<0.05).Theanastomosis of lymphatic vessels and vascular in human breast cancer axillary metastaticlymph nodes was16.7%.3.There were no statistical significance in lung metastatic rates in Walker-256metastatic lymph node.Efferent lymphatics ligation group and non-ligation group wasrespectively70%and90%(p=0.582).4.Walker-256pelma translanted tumors in rats, the group resecting foot lesions byamputation and ligating the vascular and lymphatic of lymph node and the group radicalresecting foot lesions by amputation and rescting lymph node, the recurrence rates wereboth0after eight weeks, had no significance. Conclusion:1.Endostar inhibit the microvessel and microlymphaticvessel endothelial genera-tion of rat Walker-256popliteal fossa metastatic lymph node, inhibit transplanted tumorspopliteal fossa lymph node metastasis.2.There is the anastomosis of lymphatic and vascular possibely in Walker-256metastatic lymph nodes and human breast cancer metastatic axillary lymph node.
