Effect Of Bisphenol A And CdCl2on The Viability Of MCF-7Cells And The Protein Expression Of WISP-2and Bcl-2 | Posted on:2013-08-12 | Degree:Master | Type:Thesis | Country:China | Candidate:M Ceng | Full Text:PDF | GTID:2234330374979264 | Subject:Health Toxicology | Abstract/Summary: | PDF Full Text Request | Objective: Cell culture was used to investigate and explain the combined effectof bisphenol A and cadmium chloride on the proliferation of MCF-7human breastcancer cells and whether BPA promote the expression of WISP-2and controlBCL-2.It also tried to explore how to influence WISP-2and BCL-2whenbisphenol A combined with CdCl2.Methods: The changes of cell were observed by light microscopy after24h withdifferent concentrations of bisphenol A and cadmium chloride role in MCF-7cells.The effects of bisphenol A and cadmium chloride on AMCF-7cell proliferationrate by MTT method. The protein expression of WISP-2and BCL-2mRNA,WISP-2and Bcl-2of bisphenol A and the combined effects of the bisphenol A andcadmium chloride was analyzed by Q-PCR and Western-blot method. Estradiolwas a positive control group and was analyzed by SPSS13.0.Results:1. BPA showed a low concentration for the inhibition and highconcentrations promote the proliferation of MCF-7cells, there was significantdifference. The proliferation of MCF-7cells was induced by BPA and reached apeak at10-6mol/L and the proliferation of MCF-7cell was inhibited with10-3mol/L concentration.2. The proliferation of MCF-7cells was induced with Cadmium chloride atcertain concentrations to simulate the estradiol, and peaked at10-6mol/L, andMCF-7cell proliferation was inhibited with increasing concentrations of10-4mol/L.3. MTT showed that MCF-7cell proliferation was promoted with thecombined exposure of Bisphenol A cadmium chloride, but lower than thevalue-added rate of bisphenol A and cadmium chloride alone and mRNA andprotein expression of WISP-2and Bcl in MCF-7cells were lower than theseparate exposure group. Conclusion:1. Bisphenol A, cadmium chloride alone at certain concentrations canpromote MCF-7cell proliferation, both alone role in promoting was higher thanthe combined effect on cell proliferation.2. WISP-2gene and BCL-2gene expression were lower than the two alonewhen the combined effects of bisphenol A and cadmium chloride on MCF-7cells at certain concentrations, there may be antagonism.3. BCL-2gene expression was controlled by WISP-2gene, bisphenol A canpromote the expression of WISP-2gene. | Keywords/Search Tags: | MCF-7, Bisphenol A, cadmium chloride, WISP-2, BCL-2, proliferation, combined effect | PDF Full Text Request | Related items |
