Objective: Establish the diabetic rat tibial bone defect model. Compare the effectsof using NGF, HA and the combination of the both on the general morphology andimaging, and the effects on calcium content and ALP in bone defect healing process.And discuss the effects and mechanisms of the combined application of NGF and HAon diabetic rat bone defects.Materials and methods758-week old healthy male SD rats were randomly divided into60diabeticgroup, receiving single dose intraperitoneal injection of50mg/kg streptozotocin(streptozotoein, STZ) and15control group, receiving intraperitoneal injection ofequal volume saline.3days after the injection, we choose60rats whose blood glucoseconcentration were higher than16.8mmol/L into the diabetic group,replenished therat which blood sugar is not reach the standard. then randomly divided them intogroup B, C, D, E with15rats in each group. We choose15rats from the control groupas the group A, whose blood sugar is normal.All groups of rats were established as4mm right upper tibia bone defect model and orderly used saline, saline, NGF, HA,NGF+HA in the bone defect. Post-operation at4th、8th、12th week, we randomlyselected5rats from each group to get venous blood sampling. Then we carried out thegeneral specimens observation, X-ray observation of bone defect, calcium content,histological observation and the detection of ALPResults: Gross observation at the same postoperative time: In diabetes group B, C,D, E,the bone tissue volume and hardness in local bone defects increasedgradually.The healing of control group A is better than group B but worse than groupD, E; X-ray showes: The transverse and longitudinal fuzzy degree of the bone callusin bone defect group B, C, D, E was increased gradually.Group A was better than group B but worse than group D, E. Histological observation: The maturity anddensity of bone trabecular in group B, C, D, E was increased.Group A is higher thangroup B but less than the remainder of the group.The content of ALP were increasedin the order of group B, C, D, E The content of serum ALP in each group reachedthe peak at8th week. Comparing to the other four groups,group E was thehighest,the difference had statistical significance(p<0.05).ALP fell at12thweek.Calcium content increased over time, increased in the order of groupB、C、A、D、E in every stage.Conclusion: NGF composited with HA has a good function inosteoinduction,activeing the growth of osteocyte,speed up the diabetes rat bonedefect healing.
