Objective: We explore the differential expressions of BMP-1in injuredMCL and ACL fibroblasts induced by TNF-α and try to find out thedifferences between the poorly self-healing ACL and well functionallyself-healing MCL fibroblasts.Methods: Biaxial loading chamber is used to injure the MCL and ACL cells,which are treated by TNF-α. The expressions of BMP-1were detected byreversed-transcript PCR and real-time quantitative PCR.Results: The results revealed that gene expression of BMP-1is lower withTNF-α treatment than control, and decreased expressions showed adose-depended manner. The expressions were down to41%in MCL and27%in ACL at24h. BMP-1expression in MCL is still higher than theexpression in the ACL, the difference of12hours up to2.02±0.18-foldConclusion: The expression of BMP-1induced by TNF-α directly affectsaxtracellular matrix synthesis, which can affect the ligament repair process.Recognizing this imply a potential significant value and clinical usage for cure of injured ACL.
