Objective:To study the influence of FTY720on the differentiation of osteoclast precursor cells (RAW264.7) induced by polyethylene particle in the co-culture system. It is used to seek a new treatment method of aseptic loosening of artificial joint.Methods:Millipore co-culture models system between osteoclast (RAW264.7) and osteoblast MC3T3were established, then FTY720was used to intervene the differentiation of cell (RAW264.7) which irritated by7-9micron diameter polyethylene wearing particle; Tumor necrosis factor-a (TNF-a) and interleukin-6(IL-6) were detected by the method of enzyme-linked immunosorbent assay (ELISA); mRNA expression of tartrate-resistant acid phosphatase (TRAP) and nuclear receptor coactivators (RANK) gene were investigated, and bone osteolysis was observed by scanning electron microscope(SEM).Result:TNF-α、 IL-6excretion and bone osteolysis were significantly increased in granules group, while they were inhibited in the group with FTY720. Conclusion:FTY720could effectively inhibit the differentiation of osteoclast precursor cells (RAW264.7), not only reduce osteoclast formation, bone osteolysis absorption and as well as reduction of the secretion of inflammatory factors(TNF-α、IL-6), down regulated the expression of the typical osteoclast phenotype gene TRAP and function gene RANK. FTY720might promise to become one of the drugs of preventing aseptic loosening of artificial joint. |
