Objective:To Preparation one of nano-targeted and wrapped131I drugs,which thyroid cancernoma cell line can be specifically targeted by TSHreceptor-mediated and to study their targeting ability in vitro.Methods: The high molecular polymer PLGA-COOH Nanoparticleswhich wrapped radionuclide131I were produced by the double emulsiontechnique.TSH antibody was covalently linked to the PLGA-COOHnanoparticle surface using a carbodiimide technique. To express theposition of TSH receptor on thyroid cancernoma cell line throughImmunofluorescence staining. To detect the general characteristics andtargeting ability of the targeting drugs in vitro. And compared with theordinary polymer nanoparticles, to exclude electrostatic adsorption.Results:Its physical property was determined. The TSH receptorexpress both membrane and cytoplasm in Thyroid cancernoma cellline(FTC-133), which was highly expressed on membrane. and thePLGA-COOH nanoparticle targeting specifity to Thyroid cancer cells wasobserved,normal Nanoparticles can be more proactive to find the target forthyroid cancer, while the conjugation in control group was negative. Conclusion: The preparation of the TSH receptor-mediated targeting ofpolymeric Nanoparticles was successful, the targeted Nanoparticles canbond to thyroid cancer cells effectively in vitro....
