Objective: To study the effect of ulinastatin(UTI) on adhesion,Invasion and Migration of estrin receptor-negative (ER-) breast cancer cellMDA-MB-231and estrin receptor-positive (ER+) MCF-7and RelevantMechanism.Methods: The breast cancer cell line MDA-MB-231and MCF-7weretreated in randomly divided4groups respectively:Control,UTI atlow,medium and high dosages. The Neutrophil elastase in both two kind ofcell culture supernates by elisa.Western Blot analysis are launched toquantify the proteins of Protein kinase B (Akt) and Integrinβ1ofMDA-MB-231and MCF-7. The adhesion ability of the two kinds cells aretested by MTT assay with Matrigel. The invasion ability of cells are testedby Transwell chamber with Matrigel. The migration of cells are examinedby Transwell chamber.Results: The expression of Akt and Integrinβ1in MDA-MB-231andMCF-7was decreased compared with the control group,while alsodecreased the of NE levels of cell culture supernates; the transfer of MDA-MB-231and MCF-7cells were inhibit compared with the controlgroup.Conclusions: UTI can inhibit the transfer of the MDA-MB-231andMCF-7cells by down regulation the Protein kinase B、Integrin B1andNeutrophil elastase. The800u/ml is the best dosages of UTI In vitro.
