Objective To investigate all-trans retinoic acid on apoptosis andproteinuria of adriamycin-induced nephropathy mice.Methods BALB/C mice(6to8wk old) were randomly divided intonormal control,ADR-induced nephropathy(model),andATRA-treated(treatment) groups.ADR nephropathy was induced by asingle tail intravenous injection of ADR(10.5mg/kg weight).The model ofADR induced nephropathy in mice (model group) was replicated,meanwhile the ADR induced nephropathy all-trans retinoic acid treatmentgroup (treated group) was designed.2wks after adriamycin injection, ATRA(dissolved in corn oil) at20mg/kg body weight was administered on3timesa week by intraperitoneal injection for10weeks, while20mg/kg corn oil3times once a week by intraperitoneal injection in control group and modelgroup. Six mice were sacrificed at6and10weeks after ADR injection. Theexpression of Fas、 FasL was measured by RT-PCR andimmunohistochemistry. The mRNA expression of RARa、RXRa detected byRT-PCR(P<0.05)。Results The level of proteinuria, TC and TG of normal group and treatment group decreased significantly(P<0.05, vs model group), and thelevel of serumalbumin increased significantly(P<0.05, vs model group).Immunohistochemistry results demonstrated Fas and FasL expression innormal group and treatment group decreased significantly(P<0.05, vs modelgroup). Fluorescent PCR results demonstrated that Fas and FasL expressionin normal group and treatment group decreased significantly(P<0.01, vsmodel group). In the10th week, mRNA expression level of ATRA receptorsRARa and RXRa of normal group and treatment group increasedsignificantly(P<0.05, vs model group).Conclusion All-trans Retinoic Acid has anti-apoptosis by Fas,FasL、on adriamycin induced nephropathy mice to protect kidney damage anddelay fibrosis process.
