Objective: To investigate the effect of down-regulation of SpecialAT-rich sequence-binding protein-1(SATB1) induced by RNAinterference (RNAi) on cell apoptosis of human lung cancer cell line A549and to explore its possible mechanism.Methods:SATB1-targeted shRNAexpression plasmid (SATB1-shRNA) was constructed.SATB1-shRNAwas transfected into A549cells by LipofectAMINE2000.3.The mRNAsand proteins expression of SATB1, Bcl2, BAX and Caspase-3wereexamined by RT-PCR and Western-Blot, respectively. The apoptosis rateof A549cells was detected by FCM. SATB1-shRNA was constructed andtransfected into A549cells successfully.Results:SATB1-shRNA werevalidated by gel electrophoresis and DNA sequence analysis. The mRNAand protein expression of SATB1was decreased markedly aftertransfection with SATB1-shRNA (P<0.05).Cell apoptosis rate inSATB1-shRNA transfection group was higher than that in the controlgroup (14.18±1.59%vs1.84±0.57%, P<0.05). The mRNA and proteinexpression of Bcl2was decreased markedly, the mRNA and proteinexpression of BAX and Caspase-3was increased after transfection withSATB1-shRNA (P<0.05).Conclusions:SATB1-shRNA candown-regulate pronouncedly SATB1expression in A549cells, induce cellapoptosis. The mechanism may be related to SATB1-shRNA reduce Bcl2 gene expression caused by the cascade effect.
