Purpose:The establishment experiment using the effect of Streptozotocin diabeticnephropathy model in rats using immUNohistochemical methods, study of plasmaurotensin Ⅱ and correlation of atherosclerosis in rats with diabetic nephropathy and itsdome chlorpromazine effect on both.Methods:Male SD rats40randomly divided into normal control group of NC (10PCs),diabetic nephropathy DN groups (15), sichuan dome chlorpromazine CZ Group (15),with a high sugar, high fat diet (ingredients: lard from10%,20%sugar, egg yolkpowder10%, composed of six bile acid sodium salt and0.5%General feed) producedAS model of diabetic rats. DN set, CZ group feeding with high sugar, high fat diet,120g/l of drinking fruit in syrup,1month after intraperitoneal injection of35mg/kg STZ(sildenafil citrate pH4.20.1mol/L prepared buffer); NC group giving general diet,injections sildenafil citrate buffer as a control. On this basis, CZ group of150mg.kgformulation of a vaulted·/d (concentration:0.1g/100ml) gavage, NC group, the DNgroup5ml.kg/d after intragastric infusion with normal saline, filling1sooner or later. At16weekend executed animal, measurement weight; left take24h urine volume, recordsand measuring24h urine protein quantitative (24h u-TP); left take blood specimensmeasuring blood glucose (GLU), and glycerol three ester (TG), and cholesterol (TC),and high density fat protein (HDL), and blood muscle anhydride (SCR), and ureanitrogen (BUN); left kidney specimens measuring kidney weight, on part kidneyorganization and abdominal active pulse line HE dyeing, in light mirror Xia observationkidney ball kidney small tube, abdominal active pulse organization morphology change;line immune group of, microo-image photography system recovery Image after j medical image analysis system, detect UⅡ, GPR14protein expression, mean averagearea of glomerular andglomerular volume.Results:1.16rats compared to the NC group DN group over the weekend, DN group rat bodyweight drop CZ group slow weight gain in rats, but more weight DN group, index ofkidney (renal/weight, KI),24h urine volume, urine protein quantitative, GLU, TG, TC,HDL, SCR and BUN all statistically difference (P<0.05).2. Light mirror check results: DN group abdominal active pulse visible bubble cell andfat sheath, smooth muscle cell deformation, CZ group more DN group light, NC groupartery wall endothelial cell light whole, arranged rules; DN group kidney visible clearkidney ball hyperplasia mast, and fiber of and structure collapse, of film matrixincreased, kidney small tube appears clear focal shaped empty bubble sample variable,CZ group more DN group light, NC group kidney ball, and kidney small tube structurenormal.3.Compared with the NC group, aorta, renal DN group UⅡ, GPR14expressionincreases, increased glomerular area, volume, UⅡ mRNA expression in renal tissueincreases (P<0.05), and DN comparison, CZ Group expressed significantly reduced(P<0.05).ConcLUsions:1.Dome chlorpromazine can to a certain extent improve the general status of diabeticnephropathy rats, capable of regulating blood sugar, blood lipid metabolism, reduceurine protein, inhibits deterioration of renal function.2. UⅡ and its receptor GPR14prompted its high expression in diabetic nephropathymay play an important role in the process of atherosclerosis. Dome chlorpromazine maybe expression by inhibiting UⅡ, and GPR14exceptions to protect kidney function.
