Objective:To investigate the effect of Total glucosides of paeony on the expression ofToll-like receptor9(TLR9) in peripheral blood B lymphocytes and the secretion ofinterferon-alpha (IFN-α) of peripheral blood mononuclear cell of systemic lupuserythematosus patients.Methods: Sixty SLE patients (study group) and thirty healthy donors(control group)were enrolled, Peripheral blood mononuclear cell (PBMC) was isolated and dividedinto5groups. In study group, PBMC of group A was the blank group, additional withnothing else, PBMC of group B was incubated with CpG-ODN, PBMC of group Cwas incubated with CpG-ODN+immunosuppressive-ODN, PBMC of group D wasincubated with CpG-ODN+Total glucosides of paeony, PBMC of group E wasincubated with Total glucosides of paeony; PBMC of control group was divided intogroup a, group b, group c, group d and group e, and incubated according to themethod above. Each group were cultured respectively for48hours. FLA (Flowcytometry analysis) was used to detect the expression of TLR9on peripheral bloodCD19+B lymphocytes. ELISA was used to detect the secretion of IFN-α insupernatants. SPSS13.0software was used to statistical analysis.Results:1. TLR9expression:①group A9.20±3.43(%), group B14.86±3.42(%),group C10.73±3.98(%), group D11.95±3.63(%), group E9.47±3.34(%), thegroup B was higher than the group A, the difference was statistical significant(p<0.001). the group C was lower than the group B, the difference was statisticalsignificant (p=0.007), the group D was lower than the group B, the difference was statistical significant (p=0.038); As to the MFI, there was no statistical differencefound between each group;②The SLE patients were divided into2groups accordingto SLEDAI score. In the slightly active group, group A5.19±2.05(%), group B10.74±3.17(%), group C5.93±1.86(%), group D7.87±2.46(%), group E5.65±2.13(%),the group B was higher than the group A, the difference was statistical significant(p=0.026). the group C was lower than the group B, the difference was statisticalsignificant (p=0.040); In the moderately/highly active group, group A10.80±2.37(%), group B16.51±1.72(%), group C12.65±2.73(%), group D13.59±2.58(%), group E11.10±2.34(%), the group B was higher than the group A, thedifference was statistical significant (p<0.001). the group C was lower than the groupB, the difference was statistical significant (p<0.001), the group D was lower than thegroup B, the difference was statistical significant (p=0.008),③In the health people,group a4.96±2.11(%), group b9.10±2.12(%), group c6.27±1.77(%), group d6.98±2.10(%), group e5.43±1.60(%), the group b was higher than the group a, thedifference was statistical significant (p=0.002). the group c was lower than the groupb, the difference was statistical significant (p=0.012), As to the MFI, the group b washigher than the group a, the difference was statistical significant (p=0.048), the groupc was higher than the group a, the difference was statistical significant (p=0.018).2.The concentration of IFN-α in the supernatants:①group A4.83±0.53(μg/ml),group B6.98±0.21(μg/ml), group C5.04±0.50(μg/ml), group D5.21±0.47(μg/ml),group E1.14±0.17(μg/ml), compared with the group B, the concentration of IFN-αin the group A and the group C was lower, the differences were statisticalsignificant(p<0.001), the group D was lower than the group B, the difference wasstatistical significant (p<0.001), The difference between of the group A and the groupE was statistical significant (p=0.004);②The SLE patients were divided into2groupsaccording to SLEDAI score, the moderately/highly active group and the slightlyactive group, consequences were similar to the SLE patients.③In the health people,group a1.42±0.21(μg/ml), group b4.72±0.64(μg/ml), group c1.64±0.22(μg/ml),group d1.91±0.18(μg/ml), group e1.14±0.17(μg/ml), compared with the group b,the concentration of IFN-α in the group a and the group c was lower, the differences were statistical significant(p<0.001), the group d was lower than the the group b, thedifference was statistical significant (p<0.001), The difference between of the group aand the group e was statistical significant (p=0.048).Conclusions: Our data indicate that CPG-ODN can upregulate the expression ofTLR9in peripheral blood B lymphocytes and the IFN-α secretion of PBMC,Immunosuppressive-ODN can downregulate the expression of TLR9in peripheralblood B lymphocytes and the IFN-α secretion of PBMC induced by CpG-ODN invitro. Total Glucosides of Paeony can downregulate the expression of TLR9inperipheral blood B lymphocytes and the IFN-α secretion of PBMC induced byCpG-ODN in vitro.
