Objective:The purpose of this project was to investigate the prostate cancer (Prostatecarcinoma) and adjacent noncancerous prostate tissue Hedgehog (Hh) signalingpathway factor Sonic of hedgehog (Shh), Patched1(Ptc1) and Gli1mRNA ofexpression and meaning, to explore Hh signaling in prostate cancer development roleand significance in the diagnosis and treatment of prostate cancer.Methods:Collected32cases of prostate cancer, adjacent noncancerous prostate tissuefresh specimens by reverse transcriptase polymerase chain reaction (reverseanscription Polymerase Chain Reaction RT-PCR) to detect32cases of prostatecancer, adjacent noncancerous prostate tissue in of Shh, Ptc1and Gli1mRNAexpression, and analysis of three in the pathogenesis of prostate cancer with clinicalstage, tumor grade, preoperative PSA, and PSA and clinical stage, pathologicalgrade of the relationship and its statistical significance.Results:(1)32cases of prostate cancer the number of positive expression of Shh, Ptc1and Gli1followed by29cases (90.6%),30cases (93.75%),32cases (100%),corresponding to cancer and adjacent normal prostate tissue followed by13cases(40.63%),14cases (43.75%),17cases (53.12%). Shh, Ptc1and Gli1expression infresh tissues of32cases of prostate cancer were higher than the adjacent normalprostate tissue was statistically significant (P <0.05).(2) Prostate cancer ShhmRNA levels compared with adjacent noncancerousprostate tissue increased2.1-fold, Ptc1mRNA levels2.3times,5.3-fold higherGli1.(3) The expression of Ptc1and Gli1in prostate cancer specimens werecorrelated, but the statistical analysis meaningless (P>0.05). MRNA expression ofShh and Gli1also have some relevance, but the statistical analysis meaningless (P> 0.05). Shh and Ptc1expression was positively correlated (P <0.01), the larger theexpression amount in ShhmRNA Ptc1expression.(4)32cases of prostate cancer specimens of Shh, Ptc1, Gli1mRNA expressionand clinical stage, histological grade was significantly correlated (P <0.05), theTNM clinical staging later the higher the Gleason score, pathological the lower thedegree of differentiation, the expression of Shh, Ptc1, Gli1is the stronger.(5) Preoperative serum PSA and prostate cancer, Shh, Ptc1, Gli1expressionand clinical stage, Gleason pathologic grade were correlated (P <0.05).Conclusion:(1) Shh, Ptc1and Gli1expression in prostate cancer tissues were higher than inadjacent normal tissue (P <0.05), abnormal activation of Hh signaling pathway inprostate cancer cells prompted this pathway may play in the pathogenesis of prostatecancer to an important role.(2) By contrast, found that Gli1expression in prostate cancer was significantlyhigher than in adjacent normal tissue (P <0.05), and Gli1compared with Shhexpression to the degree of tumor differentiation in more significant, also shows thatGli1can more accurately reflect the level of cancer cell proliferation anddifferentiation.(3)Serum PSA for prostate cancer patients with clinical stage and histologicalgrade positive correlation, suggesting that the degree of malignancy and prognosis ofprostate cancer by serum PSA may be a preliminary estimate.(4) Actively explore the impact of the Hh signaling pathway in prostate cancer,looking for more effective Hh pathway antagonists, further open new avenues for theprostate cancer drug treatment, diagnosis and treatment of prostate cancer hasfar-reaching significance.
