Objective:To compare the changes of cytokines IL-2, IL-10and TGF-β1inperipheral blood serum and FOXP3mRNA expression in peripheral bloodmononuclear cells TACE before and after treatment in patients with liver cancer, inorder to investigate TACE treating liver cancer may immunological mechanism.Methods: Peripheral blood samples of30cases of HCC patients before TACEchemotherapy、after chemotherapy one week later,14cases of healthy people and14cases of patients with chronic hepatitis B were collected. Measured the content ofIL-10、IL2and TGF-β1by ELISA, detected the FOXP3mRNA expression byRT-PCR with GAPDH as an internal reference.Results:①ELISA results showed that IL-2level in HCC patients before TACEchemotherapy was lower than chronic hepatitis B group and healthy people group, thedifference was statistically significant(P<0.01); IL-2level in chronic hepatitis Bgroup was lower than in healthy people group, the difference was statisticallysignificant(P<0.01); IL-2level increased in HCC patients after TACE chemotherapy,the difference was statistically significant(t=31.028, P<0.01).②IL-10level in HCC patients before TACE chemotherapy was higher thanchronic hepatitis B group and healthy people group, the difference was statisticallysignificant(P<0.01); IL-10level in chronic hepatitis B group was higher than inhealthy people group, the difference was statistically significant(P<0.01); IL-10leveldecreased in HCC patients after TACE chemotherapy, the difference was statisticallysignificant(t=51.266, P<0.01).③TGF-β1level in HCC patients before TACE chemotherapy was higher thanchronic hepatitis B group and healthy people group, the difference was statisticallysignificant(P<0.01); TGF-β1level in chronic hepatitis B group was higher than inhealthy people group, the difference was statistically significant(P<0.01); TGF-β1level decreased in HCC patients after TACE chemotherapy, the difference wasstatistically significant(t=64.812, P<0.01). ④One-way ANOVA results suggested that FOXP3mRNA expression differencewas statistically significant among the healthy people control group, the chronichepatitis B group and before TACE chemotherapy group(P<0.01); LSD resultsshowed that FOXP3mRNA expression level in healthy people was lower thanchronic hepatitis B group and before TACE chemotherapy group, the difference wasstatistically significant(P<0.01); FOXP3mRNA expression level in chronic hepatitisB group was lower than before TACE chemotherapy group, the difference wasstatistically significant(P<0.01); FOXP3mRNA expression level decreased in HCCpatients after TACE chemotherapy, the difference was statistically significant(t=48.3,P<0.01).⑤By linear correlation analysis, it showed that FOXP3mRNA expressionchanges were associated with the cytokines IL-2, IL-10and TGF-β1concentrationchanges(P<0.01).Conclusion:①FOXP3mRNA expression in peripheral blood mononuclear cells in HCCpatients was higher than in patients with chronic hepatitis B and healthy people.FOXP3is the specific marker of Treg cells, it indicates that regulatory T cells (Treg)relate to the occurrence and development of liver cancer.②FOXP3mRNA expression level decreased in HCC patients after TACEchemotherapy, it shows that chemotherapy for HCC could affect immune function.③FOXP3mRNA expression changes were associated with IL-2, IL-10andTGF-β1concentration changes by linear correlation analysis, it prompted thatregulatory T cells may play their roles in immunoregulation by IL-2、IL-10andTGF-β1.
