Objective:To establishe the SD rat model of chronic nonbacterial prostatitis (CNP) byProstatic injection of complete Freund’s adjuvant (CFA). To investigate theexpression of brain natriuretic peptide (BNP) and brain natriuretic peptide receptor(NPR-A) in the spinal dorsal horn ganglia (DRG), and the relationship between BNP,NPR-A and CNP-induced chronic neuropathic pain.Methods:1.40kept clean SD rats were randomly divided into four groups respectively,the control group, the CFA injection3-d (3-d model) group, model7-d group, model10-d group,(n=10). Injection CFA0.1ml into the prostate to establishe of CNP ratmodels;2. Extract in each experimental group rats spinal dorsal horn ganglion ofL5-S2and the prostate tissue;3. Each rat prostate tissue is divided into two parts, one part of tissue didbacteriological cultures, which culture negative take into zhe group, to excludebacterial prostatitis, another part of the prostate tissue biopsy. Routine HE staining toobserve the change of each experimental groups’ prostate tissue glands, bloodvessels, interstitial. To determine whether the model of CNP is success4. Extract the L5-S2ganglion’s RNA and transcribe it into cDNA. Then,real-time PCR, measure the expression of BNP and NPR-A in each group, explorethe relationship between BNP, NPR-A and non-bacterial prostatitis.Results:1. Prostate tissue bacteriological culture results were all negative, the CFA caninduced prostatitis, and produce a stable and reliable model of the rat CNP. With HEstaining, the extension of the modeling time, higher degree of inflammation inprostatea. And stable in7-d, produce consistent the type of pathology to clinical pathology. But control group has no significant inflammation;2. Real-Time PCR results showed that the BNP in the modeling,3-d,7-d and10-d group expression levels were2.16±0.35fold,1.61±0.21fold and1.32±0.36fold of the control group. Compare to control group,3-d and7-d group havesignificant difference (P <0.05);3. Real-Time PCR results showed that NPR-A in the modeling,3-d,7-d and10-d group expression levels were2.75±0.06fold,2.15±0.15fold and1.04±0.13fold of the control group. Compare to control group,3-d and7-d group havesignificant difference (P <0.05).Conclusions:CFA injection produces a stable CNP rat model. BNP and NPR-A are expressedin the L5-S2of DRGs of SD rats, and upregulation by inflammatory in prostatitis.BNP and NPR-A may be involved in the mechanism of the pain caused by CNP.
