| Objective:â‘ T o observe the clinical effective and safety of shuyusan in treatment ofmajeor depression patient, and evaluate the response to treatment for depression patient.â‘¡To observe anti depression activity of shuyusan by animal behavior test, as well asHPA axis and GR on rats caused by chronic mild unpredictable stress-induceddepression(CUMS).Methods:â‘ 187patients with depression, are randomly breaked into3groups:shuyusan groupã€Prozac group and modified Xiaoyao Power(SXP)group. The therapygroup was treated with shuyusan for one does twice a day. The western medicinecontrol group was treated with Prozac for20mg everyday. The Chinese medicinecontrol group was treated with SXP for one does twice a day. The total therapeuticcourse of the three groups was6weeks. The efficacy indexs (TCM scores, HAMDscores, SDS scores) and safety indexs (TESS scores) were been observed.â‘¡60SD ratswere randomly assigned to six groups: normal groupã€model groupã€Prozac groupã€high-dose shuyusan groupã€low-dose shuyusan groupã€SXP group. Model rats accept the21-day unpredictable stress stimulate, the anti depression activity will be evaluated bythe open-field test and sugar consumption test during0,3,6weeks.The level of CRHã€ACTHã€CORT in rat serum and the level of glucocorticoid receptor exprssion(GR)inhippocampus were measured.Results:â‘ The HAMD scores and SDS scores show that shuyusan group has goodeffective after3weeks treatment, shuyusan and Prozac could obvious improve sypotomand reduce HAMD scores and SDS scores,which has significantly decreased aftertreatment (P<0.05). There was no significant difference between shuyusan group andProzac group (P>0.05), but for side-effect in shuyusan group was lesser than that inProzac group (p<0.05).â‘¡After21d chronic unpredictable stress stimuli, the horizontalmovement and vertical movement index show significantly changed. The index ofhorizontal and vertical movement in mode group was significantly reduced than normalcontorl group (P<0.05). After3weeks treatment, the index of horizontal and vertical movement in High-dose shuyusan group and fluoxetine group were significantlyincreased compared with the model group (P<0.05). The immobility time in forcedswimming test for high-dose shuyusan group and Prozac group were significantlyimproved(p<0.05)The level of ACTH and CORT in high-dose shuyusan group weresignificantly decreased compared with the model group (P<0.05) and the level of CRHin Prozac group was significantly decreased compared with the model group (P<0.05).Conclusions:â‘ Shuyusan has effective to treatment depression patient, it couldimprove the symptom of TCM scores, reduce HAMD scores and SDS scores. Shuyusanhas less adverse reaction and high safety factor, so it is worth using in the clinical.â‘¡T heanimal behavial result indicated that Shuyusan had antidepressant activity effecton chronic stress-induced depression model rats. The potential antidepressant effect maybe related to decrease the level of ACTH and CORT, regulate the function of HPA axisand increae glucocorticoid receptor exprssion in hippocampus on chronic stress-induceddepression model rats. |
