Objective and significanceTo observe the effects of allogenous erythrocytes on cytokines of IL-8、IL-10、IL-12in immunological reactions activated by antigens by means of natural isolatedexperimental system.MethodsSuspension of erythrocytes,suspension of leucocytes and plasma are separated fromfresh anticoagulated homotype whole blood of healthy adults by routine method. Usinghuman allogenous erythrocytes as activating antigen (using autogenous erythrocytes ascomparison)and autologous plasma as reaction substratum, we investigate the effects ofallogenous erythrocytes on cytokines IL-8、IL-10、IL-12. After incubation an hour in37℃,supernate fluid is suspended and detected the change of IL-8、IL-10、IL-12by ELISA assay.Meanwhile,we discuss the significance on the alterations.ResultsThe level of IL-8⑴There’s significant difference between the level of IL-8allogenous erythrocytesphysiological saline experimental system group [28.89179±2.674565pg/ml] andallogenous erythrocytes human Escherichia coli experimental system group[119.05455±31.056229pg/ml] and it’s difference is [-90.1628±59.37619pg/ml],(p=0.005<0.05).⑵There’s significant difference between the level of IL-8autologous erythrocytesphysiological saline experimental system group [29.05148±2.972132pg/ml] andautologous erythrocytes human Escherichia coli experimental systemgroup[132.63303±35.072837pg/ml]and it’s difference is [-103.5816±59.37619pg/ml](p=0.000<0.05).⑶There’s no significant difference between the level of IL-8allogenous erythrocytesphysiological saline experimental system group and autologous erythrocytes physiologicalsaline experimental system group it’s difference is [-0.1599±59.37619pg/ml],(p=0.819>0.05).⑷There’s no significant difference between the level of IL-8allogenous erythrocyteshuman Escherichia coli experimental system group and autologous erythrocytes human Escherichia coli experimental system group,it’s difference is [-13.5785±59.37619pg/ml](p=0.997>0.05).The level of IL-10⑴There’s no significant difference between the level of IL-10allogenous erythrocytesphysiological saline experimental system group [27.90894±5.445463pg/ml] andallogenous erythrocytes human Escherichia coli experimental system group[22.28497±4.010929pg/ml] and it’s difference is [5.6240±5.37453pg/ml](p=0.296>0.05).⑵There’s no significant difference between the level of IL-10autologous erythrocytesphysiological saline experimental system group [24.15185±3.748592pg/ml] andautologous erythrocytes human Escherichia coli experimental system group[27.58494±4.740832pg/ml] and it’s difference is [-3.4331±5.37453pg/ml(]p=0.524>0.05).⑶There’s significant difference between the level of IL-10allogenous erythrocytesphysiological saline experimental system group and autologous erythrocytes physiologicalsaline experimental system group, it’s difference is [3.7571±5.37453pg/ml](p=0.325>0.05).⑷There’s significant difference between the level of IL-10allogenous erythrocyteshuman Escherichia coli experimental system group and autologous erythrocytes humanEscherichia coli experimental system group,it’s difference is [-5.2999±5.37453pg/ml](p=0.485>0.05).The level of IL-12⑴There’s no significant difference between the level of IL-12allogenous erythrocytesphysiological saline experimental system group [25.80663±3.872463pg/ml] andallogenous erythrocytes human Escherichia coli experimental system group[22.97730±2.298994pg/ml] and it’s difference is [2.8289±4.48095pg/ml](p=0.528>0.05).⑵There’s no significant difference between the level of IL-12autologous erythrocytesphysiological saline experimental system group [37.74982±5.783372pg/ml] andautologous erythrocytes human Escherichia coli experimental system group[33.32391±2.982255pg/ml] and it’s difference is [4.4259±4.48095pg/ml (]p=0.324>0.05).⑶There’s significant difference between the level of IL-12allogenous erythrocytesphysiological saline experimental system group and autologous erythrocytes physiologicalsaline experimental system group, it’s difference is [-11.9436±4.48095pg/ml] (p=0.008<0.05).⑷There’s significant difference between the level of IL-12allogenous erythrocyteshuman Escherichia coli experimental system group and autologous erythrocytes humanEscherichia coli experimental system group, it’s difference is [-10.3466±4.48095pg/ml],(p=0.022<0.05).ConclusionsIn the whole blood isolated experimental system, allogenous and autologouserythrocytes have no significant effects on cytokines of IL-8, IL-10,this indicates thatallogenous erythrocytes have no power to modify the excretion of Macrophages-Monocytes and Th2cells; The level of IL-12of allogenous erythrocytes group is muchlower than autologous group,it indicates that allogenous erythrocytes have the ability todown-regulate excretion of IL-12of DC cells and Macrophages.Objective and significanceTo observe the effects of allogenous Leukocytes on cytokines of IL-8、IL-10、IL-12in immunological reactions activated by antigens by means of natural isolatedexperimental system.MethodsSuspension of erythrocytes,suspension of leucocytes and plasma are separated fromfresh anticoagulated homotype whole blood of healthy adults by routine method. The level of IL-12⑴There’s no significant difference between the level of IL-12allogenous leukocytesphysiological saline experimental system group [47.30961±8.117041pg/ml]and allogenousleukocytes human Escherichia coli experimental system group[39.69006±6.517480pg/ml]and it’s difference is [7.6402±4.48095pg/ml](p=0.665>0.05).⑵There’s significant difference between the level of IL-12autologous leukocytesphysiological saline experimental system group[39.95673±4.607648pg/ml] and autologousleukocytes human Escherichia coli experimental system group [29.00088±3.838735pg/ml]and it’s difference is [10.9558±4.48095pg/ml](p=0.015<0.05).⑶There’s no significant difference between the level of IL-12allogenous leukocytesphysiological saline experimental system group and autologous leukocytes physiologicalsaline experimental system group, it’s difference is [7.3529±4.48095pg/ml](p=0.102>0.05).⑷There’s significant difference between the level of IL-12allogenous leukocyteshuman Escherichia coli experimental system group and autologous leukocytes humanEscherichia coli experimental system group, it’s difference is [10.6892±4.48095pg/ml](p=0.018<0.05).ConclusionsIn the whole blood isolated experimental system, the levels of IL-8、IL-10ofautologous Leukocytes group are much higher than those of allogenous Leukocytesgroup,which indicates that allogenous Leukocytes have power to down-regulate theexcretion of some kinds of Macrophages-Monocytes,Th2cells; as the conclusions ofpart-one’s experimental study that the excretion levels of IL-8are related with IL-10,which means the two cytokines are related with each other to stabilize the equilibrium ofcytokines’ network; The level of IL-12of allogenous Leukocytes group is much lower thanautologous group,it indicates that allogenous Leukocytes have the ability to down-regulateexcretion of some kinds of DC cells and Macrophages.
