As everyone knows, with the rapid development of human society and progress ofhuman civilization, our health is facing more and more threats of non-infectious diseases.The quantities of diabetics especially type2diabetic patients are rising quickly, bringinghuge burden on the society and family.Therefore, prevention and treatment of patientswith diabetes is very important. At present, Chinese medicine treatment of diabetes isbroad. A variety of treatment of diabetes with Traditional Chinese Medicine(TCM) can notonly lower blood glucose, but also do helpful to the whole body. The composition of Jinlida particles is ginseng, yellow finish, Atractylodes, Radix Sophorae Flavescentis, RadixOphiopogonis (fry), digitalis, Radix Polygoni Multiflori, dogwood, tuckahoe, Perrin,Coptis, Zhimu, Epimedium (SSN) and Salvia miltiorrhiza. The main function issupplementing qi and nourishing yin, tonifying spleen and transporting body fluid,treatingthe deficiency syndrome of qi and yin. This study was designed to observe thehypoglycemic effect of different dose Jin lida on the diabetic rats and the protective effecton islet β cells,provide theoretical basis for clinical treatment.The experiment observed after administration of the normal control group, modelcontrol group, Jin lida of low/middle/high dose group, alpha lipoic acid group, insulingroup, metformin group rats weight, fasting plasma glucose (FBG), insulin (FINS),glycosylated hemoglobin (HbA1c), oxidative stress and inflammatory reaction (SOD,GSH, MDA, IL-1, TNF-a) index, hematoxylin eosin staining(HE staining) andimmunohistochemistry of insulin to discovery that the level of FBG,HbA1c,FINS,Weightwere decreased in high dose Jin lida group, alpha lipoic acid group,insulingroup,metformin group and the level of SOD,GSH were increased(P<0.01) while the levelof IL-1,MDA were decreased(P<0.01) in the high dose Jin lida group.The results ofpancreatic HE staining and immunohistochemistry suggested that high dose Jin lida grouphas a good repair effect on islet βcell injury.All the results show that Jinlida particles cansignificantly decrease FBG and HbA1c,modify insulin resistance,reduceinflammation,improve antioxidant,protect the function of islet β cells.The effect ofantioxidant and anti-inflammatory maybe play an important role in protecting islet β cells.Objective:To investigate the protective effect of Jin lida on isletβcells in type2diabetesmellitus rats and the probable mechanism of the protective effect. Methods:Object of the study: SPF Male Sprapue-Dawley(SD) rats. All of the rats were randomlydivided into normal control group(n=12), model control group(n=10), Jin lida oflow(n=10)/middle(n=10)/high(n=10) group, alpha lipoic acid group(n=10),insulingroup(n=10),metformin group(n=10).The first section:Every group was given drugs intragastrically. After two months,thechanges regarding weight, fasting blood glucose(FBG),fasting insulin(FINS),glycosylatedhemoglobin(HbA1c) and pathological changes in β-Islet-cells were observed by HEstaining and immunohistochemistry of insulin.The second section: Test of the index of inflammatory reaction and oxidativestress,including the content of SOD、GSH、MDA、IL-1'、TNF-a.Results:1.General condition of the rats: Normal control group of rats were in good condition withfavourable mental state, free activities, shiny hair, normal diet, normal stool and nodeath.The general condition of the rats in model group were poor with bad mentalstatus,lightless shaggy. The symptom of polydipsia,polyuria,polyphagia and weightloss appeard in the group. Drug groups have different degrees of improvement.2.Weight and serological examination: Compared with the normal control group, thelevel of FBG、HbA1c in model control group were significantly increased.The level ofHbA1c in the model control group were higher contract with the Jin lida of high dosegroup,alpha lipoic acid group,insulin group and metformin group(P<0.01).Comparedwith the model control group,the level of the FBG in the Jin lida of high dose group,insulin group and metformin group(P<0.01).The level of the weight were lower in themodel control group contract with the mentioned above three groups(P<0.05).3.HE staining of pancreatic tissues:Under microscope,islet is rich with fasoshape,abundant cytoplasm,integrate morphology,central nuclear and clearly boundariesin the normal control group.On the contrary,the islet is poor with irregular morphologyand edges,eccentric nuclear and increased cell volume in the model control group.Theresults were obviously improved in the Jin lida of high dose group,insulin group andmetformin group.4.Immunohistochemical staining results: Positive cells were stained into brown.Compared with normal control group,the expression of insulin is decreased while it was improved in different degrees in the other drug groups.5.Effects on the content of SOD,GSH,MDA,IL-1,TNF-a: Compared with the normalcontrol group,the content of SOD and GSH is obviously lower(P<0.01) and the contentof IL-1,TNF-a,MDA is significantly higher(P<0.01) in the model control group.Thelevel of SOD、GSH were higher(P<0.01) while the level of MDA、IL-1β were lower(P<0.01) in the high dose of Jin lida rather than those in the model control group.Conclusion:Jin lida has protective effect on the STZ-induced islet injury in T2DM ratsprobably because of reducing inflammatory reaction and oxidative stress.
