Objective: By detecting the expression of Nrf2in ovarian cancer tissues and theimpact of FSH on ovarian cancer cell reactive oxygen species, Nrf2expression regulation,and to explore whether FSH regulates the expression of Nrf2by reactive oxygen species(Reactive oxygen species, ROS) pathway, thereby promoting tumor.Methods:①The expression of Nrf2in ovarian cancer tissues was detected byimmunohistochemical methods.②FSH to the influence of the Nrf2expression in ovarian cancer cells wereall examined by Western blotting analysis.③H2O2to the influence of the Nrf2expression in ovarian cancer cells wereall examined by Western blotting analysis.④The influence of FSH in the ROS activity was evaluated by reactiveoxygen species assay kit.⑤NAC(ROS inhibitors) to the influence of the Nrf2expression(H2O2-induced) in ovarian cancer cells were all examined by Western blotting analysis.⑥NAC(ROS inhibitors) to the influence of the Nrf2expression(FSH-induced) in ovarian cancer cells were all examined by Western blotting analysis.Results:①Nrf2in ovarian cancer was highly expressed.②FSH in a dose-dependent manner promote the expression of Nrf2protein in ovarian cancer cells.③FSH in a time-dependent manner promote the expression of Nrf2protein in ovarian cancer cells.④FSH promote the generation of reactive oxygen species in ovarian cancercells.⑤Reactive oxygen species induced Nrf2protein expression in ovariancancer cells.⑥Reactive oxygen species inhibitors inhibit the expression of Nrf2proteininduced by reactive oxygen species.⑦Reactive oxygen species inhibitors inhibit the expression of Nrf2proteininduced by follicle stimulating hormone.Conclusions: The experiment discussed the regulating mechanisms of epithelialovarian cancer by ROS-medicated antioxidative defense system with the core of Nrf2signal pathway, and gets several conclusions as followed. by the different treatment on epithelial ovarian cancer cell lines Hey, detected the Nrf2protein expression in thedifferent induction of follicle-stimulating hormone, reactive oxygen species and reactiveoxygen species inhibitors. The follicle-stimulating hormone regulate the Nrf2pathwaymay be via ROS pathway. These findings will provide important experiment evidence forexploring the pathogenesis of epithelial ovarian cancer, and also provide new clues for thetreatment of ovarian cancer.
