| hemorrhagic shock still has high mortality recently. researchs show that inducedhypothermia have the protective effects in hemorrhagic shock,its mechanism may be thatcan store the cell’s energy. there has a study show that increase the content of NAD+caninhabit the oncosis related with PARP,but there was few report if induced hypothermia candecrease the tissues oncosis in hemorrhagic shock.PARP over-activation is one ofimportant factors leading to cell oncosis.in this study,we establish the volume-controlledhemorrhagic shock model which consisted of three phases,to research the effect of inducedhypothermia with PARP in hemorrhagic shock of rats.ObjectiveTo observe the effects of induced hypothermia on ATPã€NAD+ã€MDAã€SOD andthe PARP immunohistochemical staining of myocardium in hemorrhagic shock in rat,toresearch the outcome of induced hypothermia on tissue energy metabolic disturbance inhemorrhagic shock of rats,and to explore the therapeutic method to cardical protect.Methods in this study, Thirty-two SD rats were randomized into4groups: normal controlgroup(Group A); normothermia shock group(Group B);hypothermia shock group(GroupC);hypothermia control group(Group D),all animals were anesthetized, intubated throughright carotid artery.Group B and Group C were initiated to volume-controlled (2.5mL/100g weight) hemorrhagic shock within15min.at time point60min(T60) Group B and GroupC resuscitationed with lactated Ringer’s solution for2hours, in Group C and Group Dsurface cooling was initiated by applying alcohol onto the abdominal skin and blowingwith an electric fan in order to reduce rectal temperature to34±0.5℃for2hours. at theend of resuscitation,rectal temperature recoverd to normal level with lamp,followedobserved for1h before animals were killed(T240).recorded arterial pressure data at T0ã€T15ã€T60ã€T180time points,at the end of T240point,animal was killed and obtainedmyocardium, immunohistochemical staining for PARP was performed, ATPã€NAD+inmyocardium were assessed by an isocratic HPLC separation with UV detection,and MDAã€SOD contents in myocardium were assessed by ELISA.Results i the ATPã€NAD+content were decreased in Group B, Group C were increasedthan Group B,but the were still lower than Group A and D;the were no difference betweenGroup A and D; the MDA content was increased in group B,and were decreased in GroupC,but the were still higher thanGroup A and D;the were no difference between Group Aand D;the SOD content were decreased in Group B, Group C were increased than Group B,but the were still lower than Group A and D, the were no difference between Group Aand D; the PARP immunohistochemical staining postive rate were highest innormothermia shock group, and higher in hypothermia shock group, the normal controlgroup and hypothermia control group were hardly no expressed.Conclusions induced hypothermia can protect the myocardial cell energy,and improvecell Metabolism,decreased myocardial damage,decreased oncosis related with PARP ofmyocardium in hemorrhagic shock of rats... |
