Objective To investigated the regulatory effects of Erxian Decoction on reproductive endocrine and immune systems of mice.Methods Dividing female mice to five groups according to randomization. Normal control: intragastric administration of equal-volume physiological saline; Positive control:intragastric administration of10.4mg/kg lentinan; Pattern(pattern of immunosuppression):peritoneal injection of100mg/kg Cyclophosphamide from day1to day3, the density is10mg/ml, once a day, and intragastric administration of equal-volume physiological saline meanwhile; Immunosuppression+positive control:Cyclophosphamide injection is the same with pattern, and intragastric administration of10.4mg/kg lentinan; Immunosuppression+Erxian decoction: Cyclophosphamide injection is the same with pattern, and intragastric administration of11.3g/kg Erxian decoction. All medicines should be used for7days except Cyclophosphamide.①Observing, recording and comparing the common animations of mice of every experimental group, in the process of raising and administration;②Weighting uterus, ovaries, thymus and spleens of mice when finished administration, and calculated organ index, which equals to weight of the organ divide weight of the body;③Taking mice thymus and ovaries when finished administration, staining with hematoxylin and eosin (HE), and micro-observing them;④Taking blood of mice when finished administration, separating blood serum, assaying E2level using radio-immunity;⑤Assaying II-1α level of blood serum using enzyme linked immunosorbent assay (ELISA);⑥Dealing with thymus, spleens, ovaries and uterus of mice using immunohistochemical method, and micro-observing them;⑦Taking spleens from mice when finished administration, detecting cell cycle using flow cytometry.Results①We discovered that, compared with mice of normal control and positive control, others were decadent, hairs fluffy, reaction clumsy and body weight descent from the first day after Cyclophosphamide (CYC) injections. There was no dead mouse through the experiment;②After molding by CYC, the uterus index of normal control is marked higher than pattern and positive control; Two treatment groups are higher than pattern, the difference is conspicuously, and there is no significant difference between the two groups; Significant variation isn’t be found between the two treatment groups and normal control. The ovaries index of pattern is notable lower than normal control and positive control after CYC injections; Two treatment groups are obviously higher than pattern, and no significant difference exit between them; The two treatment groups show no significant variation between normal control. The spleen index of pattern group was significantly lower than that of normal control after CYC injections; The thymus index of pattern group was obviously lower than that of normal control; Erxian Decoction treatment was between them; The spleen index of Erxian Decoction treatment was noticeably higher than that of pattern group, and lower than that of normal control;③Thymus of normal control were satiate and fat, with thinner cortex, thicker marrow and abundant germinal centers; Thymus of pattern group were slender and tiny, with conspicuously shrink marrow, thicker cortex, thinner marrow and noticeably decreased germinal centers. These symptoms all improved after medication, in which the effect of Erxian Decoction was more conspicuous; In ovary observation pattern group, irregular shapes of follicles, markedly reduced primordial follicles and increased atretic follicles were presented. In Erxian Decoction treatment group, the morphology of follicles was normal, more primordial and growing follicles, some closed atretic follicles and no mature ones were seen;④The E2level of pattern group was lower than that in normal control, the E2level of Erxian Decoction was higher than that in pattern group, that’s all notable. And there wasn’t significant difference between Erxian Decoction and normal control;⑤The IL-1α level of pattern group was obviously lower than that in normal control. Compared with pattern group, two treatment groups showed no significant differences, but markedly lower than normal control;⑥ER distribute equally and widely in thymus gland of normal control, mostly in mesenchyme; The distribution of pattern is evident disorder, atrophic, long and thin, mostly in cortex; It is improved in two treatment groups, ER distribution of positive treatment group is plump, Erxian decoction group is uniform and spread. In spleens of normal control, ER scatter equally and widely, the shape is mostly full granulate; Pattern is lack of equality and full, it scatter widely; The improve of two treatment groups is not notable, they distribute a little clutter, atrophic and widely; The IL-1R distribute equally, widely and plump in ovaries of normal control, mostly in mesenchyme; Pattern is disorderly, unsystematic, auantic and contractible; Erxian decoction improves a lot. it scatters plump, widely and a bit disorder. It is hard to find IL-1R in uterus, just a small amount in uterus wall of normal control and Erxian decoction, there is no distribution in uterus of pattern;⑦The percentage of G1period shows no significant difference between normal control and pattern; S period percentage of pattern is obviously lower than normal control; The G2percentage of pattern is notable higher than normal control; The spleen cell apoptosis rate of pattern is significant higher than normal control; The apoptosis rate of Erxian decoction is distinct lower than pattern, and shows no significant difference with normal control after treatment.Conclusion Erxian Decoction has better effects on protecting immunization and regulating reproductive endocrine, and there were inter-regulations between the two systems.
