Objective:To observe the effect of Rhein on rats with chronic allograft nephropathy model of kidney lesion in rats and elucidate its mechanism of action.Methods:the F344inbred rats and Lewis inbred rats (all male) as a donor,receptor,e stablishment of a rat model of chronic allograft nephropathy.30transplant rats were rando mly divided into two groups,16in the model group (the blank group),14in the experi mental group (i.e.,rhubarb treatment group);the other5Lewis rats underwent right nephr ectomy,as compared to the control group.Collection of rat blood samples respectively afte r renal transplantation in four weeks,eight weeks and sixteen weeks,detection of the renal function in rats.The rats are Respectively killed in eight weeks and sixteen weeks after renal transplantation,production section of the rat kidney tissue,observation group of rat re nal allograft tissue pathological changes,in the further observation of rhein on animal mo del of group the following indexes,and investigate the mechanisms of drug action:1,on th e extracellular matrix such as collagen Ⅰ,Ⅲ and fibronectin effect.2,the fibrogenic cytokin es such as transforming growth factor beta1,connective tissue growth factor expression i nfluence;Results:1,compared with the model group,rhubarb acid in the treatment of rats’renal function improved;renal histopathological examination showed renal interstitial fibrosis an d interstitial inflammation has been significantly improved;2,compared with the model gro up,rhubarb acid treated group of rats renal tissue fibrosis caused by factor CTGF express ion decrease;extracellular matrix FN were markedly reduced.Conclusion:1,Rhein can improve CAN rat kidney function and chronic renal disease;2,Rhein has probably through inhibition of inflammatory cell infiltration,reduce profibrotic cytokine CTGF expression,ultimately affect the synthesis of extracellular matrix,to attenuat e renal fibrosis effect.
