| Objective:to investigate the activation of autophagy after intracerebral hemorrhage and the changes of autophagy after "nao xue tong" be applied,then combined with the neurological function sorces of the rats, discuss the link between autophagy and the nerve protection.Methods:60SD male rats were randomly divided into three groups(N=20):model group (autologous arterial blood), the control group (physiological saline) and the traditional chinese medicine group(autologous blood adds the chinese medicine by using stomach tube),and each group was randomly divided into four time points(n=5):6h,12h,24h,72h..Model group and chinese medicine group were established by injecting50ul autologous arterial blood into caudate nucleus,the control group were established by50ul physiological saline,chinese traditional medicine group were administrated by stomach5days in advance,0.5ml/100g,2times/d,after modeling,continue giving medicine according to the original way.Then at each time point every group rats were graded according to the Longa rating standards and then the specimens were taken, all specimens were stained with immunofluorescence and detection of LC3-positive number.Resluts:After intracerebral hemorrha6h, autophagy appears,12h-24h reached the peak (bcompared with6h and72h groups had statistically significant differences, P＜0.05),72h began to decline; chinese medicine group significantly reduced the number of autophagy at6h and72h both time points(compared to the model group had statistically significant difference, P＜0.05);"nao xue tong" improve the neurological score at24h and72h time points (compared with model group had significant statistical difference, P＜0.05); at point72h,"nao xue tong" maybe inhibit autophagy to improve neurological function, but it seems that more evidence is needed to determine the llink between the improvement of neurological function and autophagy reducing. |
